摘要
目的研究重症急性胰腺炎(SAP)早期心肌损害与心肌细胞凋亡的关系和意义。方法应用5%牛磺胆酸钠逆行胆胰管注射诱发大鼠SAP模型。实验设定对照组(n=18)和SAP组(n=18),并分为6、12、24h三个时点,分别检测血清LDH、CK、CK-MB,用TUNEL法检测心肌细胞凋亡。结果SAP组在各时点较对照组血清LDH、CK和CK-MB显著升高(P<0.01);对照组在各时点未见心肌细胞凋亡或仅出现极少数心肌细胞凋亡,SAP组出现较多的心肌细胞凋亡,其凋亡指数(AI)较对照组显著升高(P<0.01)。结论SAP早期可诱发心肌损害,与心肌细胞凋亡有关。
Objective To study the relationship between myocardical injury and apoptosis of cardiac myocytes induced by early severe acute pancreatitis (SAP) in rats. Methods The SAP model was induced by retrograde injection of 5% sodium taurocholate into the pancreatic duct. The rats were randomly divided into control group and SAP group. The animals were sacrificed at 6, 12 and 24 hours after operation under aseptic circumstances. The severity of injury to cardiac myocytes was determined by plasma LDH, CK, and CK-MB levels. The apoptosis of cardiac myocytes was detected by in situ TdT-mediated dUPT nick end labeling (TUNED technique. Results Plasma levels of LDH, CK and CK-MB, and apoptosis index (AI) were significantly increased in SAP group. Conclusions Early SAP can induce myocardial injury, which might be related to the apoptosis of cardiac myocytes.
