摘要
目的 :探讨大蒜新素 (即大蒜素注射液 )对鼠巨细胞病毒 (MCMV)心肌炎的治疗作用。方法 :建立鼠巨细胞病毒性心肌炎小鼠模型 ,3 0只模型鼠被随机分成大蒜新素治疗组 (n =10 )、更昔洛韦治疗组 (n=10 )和对照组 (n =10 )。以MCMVK181接种之日为 0 ,第 1天开始分别运用大蒜新素和更昔洛韦腹腔注射 ,每天 1次 ,持续 6天。对照组用生理盐水治疗对照。各组分别于第 7天和第 3 5天分别处死 5只小鼠 ,取心脏 ,心脏沿正中线横断切片 ,观察各组小鼠心肌细胞浸润炎性病灶数量 ;取脾、肝和唾液腺组织匀浆 ,用蚀斑形成实验方法进行病毒滴定 ,以观察大蒜新素对MCMV感染小鼠心肌炎疗效 ,并与更昔洛韦比较。结果 :鼠巨细胞病毒 (MCMV)感染BALB/c小鼠能导致心肌炎性细胞浸润灶形成。在心肌炎急性阶段 ,大蒜新素和更昔洛韦与安慰剂组比较 ,能显著抑制模型鼠心脏浸润炎性病灶数量和脾、肝和唾液腺组织中病毒滴度( P <0 .0 1) ,且两药疗效无明显性差异 ( P <0 .0 1)。但在心肌炎慢性阶段大蒜新素疗效与更昔洛韦比较有明显差异 (P >0 .0 5 )。结论 :鼠巨细胞病毒 (MCMV)感染BALB/c小鼠能导致心肌炎的发生 ;在心肌炎急性阶段 ,大蒜新素能有效地减少心肌细胞浸润炎性病灶数量和抑制脾、肝和唾液腺组织中病毒复制 ,其治疗效果与?
Objective:To investigate the effects of Allitridin on the murine cytomegalovirus-induced myocarditis.Methods:A BALB/c mice model system of murine cytomegalovirus-induced myocarditis was established.30 mice were allocated randomly into Allitridin injected group (n=10),Ganciclovir injected group (n=10) and blank controlled group(n=10).Mice were infected with MCMV on day 0.Drug treatments commenced on day 1 post infection (p.i.) and continued daily.25mg/kg.d of Allitridin,40mg/kg.d of Ganciclovir was used by intraperitoneal(IP)injection(once per day 6 days).Saline was similarly administered as a placebo.Hearts,livers,spleens,and salivary glands were removed from groups of five mice on day 7and day 35 p.i.The hearts were transected along the midline and processed with formaldehyde fixative for histological examination.The spleens,livers,and salivary glands were homogenized,and infectious virus was quantified by plaque assay.Results:In studies examining the acute phase of myocarditis,MCMV-infected mice treated with Allitridin showed a significant(P< 0.01)reduction in virus titers in the livers,spleens,salivary gland and myocarditis compared to the untreated MCMV-infected mice and no significant difference compared with Ganciclovir treate group(P>0.05).While in experiments examining the chronic phase of myocarditis,the effects of Allitridin showed a significant(P<0.05)difference in virus titers compared to Ganciclovir.Conclusion:MCMV infection could lead to myocarditis in mice.Allitridin can obviously reduce MCMV titers,its efficacy was similar to that of Ganciclovir in the acute phase of myocarditis.
