1α,25-二羟维生素D_3和中波紫外线对正常人黑素细胞增殖及黑素合成的影响 被引量:7

Effects of 1α,25-dihydroxyvitamin D_3 and UVB on cell proliferation and melanin synthesis of cultured human melanocyte

摘要 目的 :探讨 1α ,2 5 二羟维生素D3 和中波紫外线 (UVB)对黑素细胞增殖及黑素合成的影响。方法 :体外黑素细胞培养 ,用四甲基偶氮唑蓝比色法 (MTT法 )测定 1α ,2 5 二羟维生素D3 和UVB(5 5mJ/cm2 )对黑素细胞增殖的作用 ;用NaOH处理后比色法测定黑素的含量。结果 :浓度为 10 -7~ 10 -10 mol/L的 1α ,2 5 二羟维生素D3 对体外培养的黑素细胞有促增殖作用 ,同时黑素含量也有增加 ,其中以 10 -7mol/L 1α ,2 5 二羟维生素D3 对黑素细胞作用最为明显。UVB照射与 1α ,2 5 二羟维生素D3 共同作用于黑素细胞时也可促进黑素细胞增殖 ,但二者无明显的协同作用。结论 :1α ,2 5 二羟维生素D3 和UVB能促进黑素细胞增殖及黑素合成 ,可用于治疗白癜风。 Objective:To study the effects of 1α,25-dihydroxyvitamin D 3 and UVB on cell proliferation and melanin synthesis of normal human melanocytes. Methods:Melanocytes of foreskins of healthy men were cultured and treated with various concentration of 1α,25-dihydroxyvitamin D 3 or UVB (55 mJ/cm 2),or both.Cell proliferation was measured with MTT assay .The synthesis of melanin was determined by chromatography. Results: 1α,25-dihydroxyvitamin D 3 and UVB could promote the proliferation of cultured melanocyte,and 1α,25-dihydroxyvitamin D 3 could promote the melanin synthesis.The significant concentration of 1α,25-dihydroxyvitamin D 3 ranging from 10 -7 to 10 -10 mol/L.Conclusion:1α,25-dihydroxyvitamin D 3 and UVB irradiation could promote the proliferation of melanocyte,which indicates that they might be effective in the treatment of vitiligo.
