神经生长因子对大鼠小脑皮质神经细胞缺氧、缺糖损伤的保护作用 被引量:1

Protective Effect of NGF on Injury Induced by Anoxia and Hypoglycemia in Cultured Rat Cerebellar Neurons

摘要 目的 探讨神经生长因子 (NGF)对小脑皮质神经细胞缺氧、缺糖损伤的影响。方法 采用含 0 5mmol·L-1连二亚硫酸钠 (Na2 S2 O4)的无糖Earle’s液复制小脑皮质神经细胞缺氧、缺糖损伤模型 ,通过形态学观察、MTT法和LDH活性的测定研究NGF对小脑皮质神经细胞的保护作用。结果 NGF (5 0 ,10 0 μg·L-1)组小脑皮质神经细胞形态损伤明显减轻 ,细胞存活率升高 ,培养液中LDH含量明显下降。结论 NGF对缺氧、缺糖引起的小脑皮质神经细胞损伤具有保护作用。 Objective To investigate the effects of NGF on injury induced by anoxia and hypoglycemia in cultured cerebellar neurons.Methods Cultured cerebellar neurons were treated with Na 2S 2O 4 and Earle's liquid with glucose free to induce models of anoxia and hypoglycemia.The protective effects of NGF were evaluated by the activity of loctate dehydrogenate(LDH) and staining with MTT assay and HE straining.Results Previously incubating with NGF(50,100μg·L -1) counteracted the injury of cultured rat cerebellar neurons induced by anoxia and hypoglycemia.Compared with control group,the contents of LDH were much lower than that in control and the neuronal viabilities were much higher in treatment groups.Conclusions NGF can protect cerebellar neurons from injury induced by anoxia and hypoglycemia.
