摘要
目的 探讨高脂血症对大鼠阴茎海绵体组织内皮型一氧化氮合酶 (eNOS)和诱生型一氧化氮合酶 (iN OS)蛋白表达的影响。方法 2 8只雄性SD大鼠随机分为实验组 (n =16 )和正常对照组 (n =12 )。实验组大鼠给予高胆固醇食料 ,正常对照组给予普通食料喂养 ,于实验开始后 6周末和 12周末 ,分别随机取各组 1/ 2大鼠观察其勃起功能 ,并采用免疫组化SP法检测阴茎海绵体组织中eNOS和iNOS蛋白质的表达情况。结果 6周末 ,实验组和正常对照组相比eNOS和iNOS的表达及阴茎勃起次数差异均有显著性 (P <0 .0 1) ,而勃起率差异不明显 ;12周末 ,两组在eNOS和iNOS蛋白的表达及阴茎勃起次数均有显著差异 (P <0 .0 1) ;实验组eNOS和iNOS的表达及阴茎勃起次数 ,12周末均明显低于 6周末 ,两两比较差异均有显著性 (P <0 .0 1)。结论 高脂血症可以明显降低阴茎组织中eNOS以及iNOS蛋白质表达 ,这可能是高脂血症引起勃起功能障碍 (ED)
Objectives To investigate the effects of hyperlipidemia on eNOS and iNOS in corpus cavernosum of rats . Methods Twenty-eight male SD rats were divided randomly into two groups ,experiment group and control group .The experiment group were fed on 1% cholesterol forage while the control group were fed on normal forage.After six weeks and twelve weeks,a half of each group were selected respectively for testing erectile function and then were sacrified for the detection of eNOS and iNOS with streptaridin-peroxidase immunohistochemistry techniques (SP method).Results Significant differences existed in erection frequency and expression of eNOS、iNOS between experiment group and control group (P<0.05) and there was also significant diffirence in expression of eNOS and iNOS between two experiment groups(P<0.05).Conclusion The expression of eNOS and iNOS in corpus cavernosum can be dramatically affected by uncontrolled hyperlipidemia.The decreasing of eNOS and iNOS expression may be one of the mechanisms underlying erectile dysfunction in hyperlipidemia subjects.
