摘要
目的 探讨5-氮胞苷对大鼠骨髓基质细胞β肾上腺素能受体(β受体)及其mRNA表达水平的影响。方法 体外分离培养大鼠骨髓基质细胞,以5-氮胞苷诱导24 h,正常培养3周,125I-Pindolol同位素放射配基法测定β受体密度,RT-PCR法测定其β受体mRNA表达水平,并与培养乳鼠心室肌细胞相比较。结果 正常培养大鼠骨髓基质细胞不表达β受体蛋白及其mRNA。5-氮胞苷诱导后的骨髓基质细胞表达β受体,随着5-氮胞苷浓度增加,骨髓基质细胞的β受体密度增加,至5×10-6mol/L时达最高峰;β受体密度随着培养时间延长而逐渐增加,于4周时接近正常心肌细胞水平。结论 5-氮胞苷诱导后,培养大鼠骨髓基质细胞表达β受体,并有明显的剂量和时间依赖性,分化后的细胞具有类似儿茶酚胺类激素的受体。
Objective To explore the effects of 5-azacytidine (5-AZ) on expression of β-adrenoceptors (β-ARs) of rat bone marrow stromal cells (BMSCs). Methods Rat BMSCs were cultured in vitro, and then were incubated with 5-AZ. Before and after treatment, 125I-Pindolol radioligand banding was used to measure B-ARs, RT-PCR was used to measure B-ARs mRNA expression in cultured rat BMSCs. Results There is no expression of B adrenergic receptors in normal cultured BMSCs. After induced by 5-AZ, the maximal number of 125 I-Pindolol banding sites in cultured BMSCs was increased markedly, B-ARs mRNA was also expressed. Conclusion The normal cultured rat BMSCs don't express B-ARs, after treated with 5-AZ, cultured rat BMSCs expressed B-ARs.
