人精子冷冻复苏后活动力和超微结构变化 被引量:1

CHANGES IN MOTILITY AND ULTRASTRUCTURE OF SPERMATOZOA AFTER FREEZE-THAW

摘要 应用超薄切片电镜技术结合精子活动轨迹摄片法研究了十二种甘油-蔗糖不同配伍的冷冻保护剂冻存精子的超微结构及活动力。结果发现:①冷冻复苏主要影响精子的膜相结构,且不同部位的生物膜对损伤的敏感性不同;②精子的活动能力明显降低;③以5%甘油、50mmol/L蔗糖配伍的保护剂效果较好。 The motility and ultrastructure of frozen-thawed human spermatozoa, which were protected by 12 different cryoprotective media composed of glycerol and sucrose, were studied by means of ultrathin-section TEM technique and photomicrograph method. The results suggested that: ①Freeze-thaw affected mainly on the plasma membrane structure, but different membranes displayed distinct sensitivity to lesions. ② Spermatozoa motility was reduced obviously. ③ Effects of 5% glycerol combined with 50mmol/L sucrose as cryoprotective medium were well.
