H5亚型高致病性禽流感疫苗的免疫效力 被引量:21

Protective Efficacy of H5 Subtype Avian Influenza Vaccines against Lethal Challenge with Homologous Virus in Commercial Chickens

摘要 利用表达H5亚型禽流感病毒血凝素基因(HA)的重组鸡痘病毒(rFPV-HA)和全病毒灭活疫苗免疫商品鸡,比较两种疫苗的保护效力。结果发现,利用103PFU和106PFU的rFPV-HA免疫无母源抗体商品鸡和含母源抗体商品鸡,HI抗体应答水平均低于灭活疫苗免疫组。利用H5亚型禽流感病毒经肌肉注射攻毒后,rFPV-HA免疫的无母源抗体商品鸡获得了完全保护(100%),高于灭活疫苗保护率(94.7%);但rFPV-HA免疫含母源抗体商品鸡诱导的保护率(11.4%~15.8%)低于灭活疫苗免疫组的保护水平(68.4%)。结果表明,rFPV-HA诱导HI抗体的能力低于灭活疫苗;但rFPV-HA免疫无母源抗体商品鸡的保护率完全达到灭活疫苗保护水平,显示出了良好的应用前景;灭活疫苗和rFPV-HA均受到母源抗体的干扰,不适于在母源抗体处于高峰期时使用。 1-day-old commercial chickens were immunized with recombinant fowlpox virus expressing haemagglutinin of H5 subtype avian influenza virus (rFPV-HA)or inactivated vaccine by subcutaneous route. Vaccinated chickens were challenged with homologous virus by intramuscular route 22 days post vaccination. The rFPV-HA induced apparently lower HI antibodies than those generated from vaccination using inactivated vaccine. However rFPV-HA provided complete protection against challenge in chickens without maternal antibodies, while inactivated vaccine induced 94.7% protection. In contrast to chickens free of maternal antibodies, rFPV-HA just induced 11.4%~15.8% protection in chickens with maternal antibodies to fowlpox virus and H5 subtype avian influenza virus, whereas inactivated vaccine provided (68.4%) protection in those chickens post challenge. The findings showed that although rFPV-HA has lower potential to induce HI antibody in vaccinated chickens compared with inactivated vaccine, but provides equal protection in chickens without maternal antibodies compared with inactivated vaccine. In addition, maternal antibodies can interfere active immunity that induced by both rFPV-HA and inactivated vaccine in chickens.
