摘要
目的 对电极法测定钾 (K)、钠 (Na)、氯 (Cl)、总二氧化碳 (TCO2 )、尿素 (Urea)、肌酐 (Cr)和葡萄糖 (Glu)等常规生化项目进行临床应用评价。方法 系统研究了电极法测定常规生化项目的线性、精密度、准确度及干扰因素 ,并分析比较与直接离子选择电极法和酶促法检测结果的相关性。结果 电极法测定K、Na、Cl、TCO2 、Urea、Cr和Glu的线性相关系数分别为 0 .992、0 .990、0 .992、0 .990、0 .997、0 .999和 0 .998。除外低值样本TCO2 的批内CV <7.4 %、批间CV <1 2 % ,其它低、中、高值样本各项目的批内和批间CV <5 %。测定K、Na、Cl、TCO2 、Urea、Cr和Glu的平均回收率分别为 96 .0 %、99.7%、95 .0 %、1 0 1 .0 %、97.0 %、96 .3%和 98.5 %。脂血和黄疸基本不影响测定结果 (P >0 .0 5 ) ,但溶血对K、维生素C对Cr测定将有显著性干扰 (P <0 .0 5 )。与直接离子选择电极法和酶促法相比较具有良好的相关性 (n =5 0 ,r >0 .933) ,结果之间的差异均无显著性意义 (P >0 .0 5 )。结论 电极法操作简便、自动化程度高、检测快速、结果准确可靠 ,具有较高的精密度 。
Objective To evaluate the analytical features of kalium(K),natrium(Na),chloride(Cl),total carbon dioxide(TCO 2),urea,creatinine(Cr) and glucose(Glu) assay using electrodes technology.Methods The parameters like linear limit,precision,accuracy and interference factor of the electrodes assay were determined.The correlation study was also performed versus direct ion selective electrodes and enzyme method.Results The linearity correlation coefficients of K,Na,Cl,TCO 2,Urea,Cr and Glu were 0.992 ,0.990,0.992,0.990,0.997,0.999 and 0.998,respectively.Except for the within run and between run CV were <7.4% and 12% for TCO 2 in plasma of low value,those of other biochem markers were <5% in plasmas of low,middle and high value.The average recovery rate of K,Na,Cl,TCO 2,Urea,Cr and Glu were 96.0%?99.7%?95.0%?101.0%?97.0%?96.3% and 98.5 %,respectively.There was no interference in electrodes method for hyperbilirubinemic and hypertriglyceridemic plasmas( P > 0.05 ),but there was significant interference for hemolyzed and vitamin C plasmas on K and Cr assay( P <0.05).The linear regression analysis showed a good correlation compared with direct ion selective electrodes and enzyme method( n =50, r >0.933),and there was no significant difference between them( P >0.05).Conclusion The electrodes method is reliable,precise,fairly rapid and easy to perform.This method is very useful and suitable for routine biochem markers determination.
