发夹式siRNA对hDNMT1基因表达的抑制作用 被引量:4

摘要 目的 研究发夹式siRNA对hDNMT1基因表达的抑制作用。方法 根据hDNMT1全长cDNA序列设计和合成与其互补的发夹式siRNA序列 ,并以发夹式siRNA序列为模板经体外合成siRNA表达盒 (SECs)。SECs通过脂质体载体转染结直肠癌细胞株HCT1 1 6后 ,经半定量RT PCR法检测hDNMT1mRNA的表达水平。结果 发夹式siRNA可以特异性地抑制hDN MT1基因。结论 通过SECs产生的发夹式siRNAs可以特异性抑制hDNMT1基因的表达 ;本研究为通过构建表达发夹式siR Objective To research the hDNMT1 gene expression inhibition of hairpin siRNA.Methods According to the gene sequence of hDNMT1 in the GenBank,we designed hairpin siRNA templates,and generated the siRNA expression cassettees.The SECs were transfected into HCT116 cells using the agent siPO RTTM XP 1.The non transfected cells were taken as a control.Inhibitory effect of siRNAs was detected by semi quantitative RT PCR.Results The hairpin si RNA generated from SECs inhibited hDNMT1 gene expression.Conclusion At the cell level,inhibition of hDNMT1 gene expression by hairpin siRNA gene rated by SECs provides basic research for inhibiting hDNMT1 gene expression study.
