慢性气道阻塞性疾病的发病机制研究:转化生长因子β1对大鼠气道重塑的影响 被引量:5

摘要 目的:观察转化生长因子β1(transforminggrowthfactor-β1,TGF-β1)对大鼠气道平滑肌细胞(airwaysmoothmusclecell,ASMC)增殖的影响。方法:ASMC取自1只健康雄性SD大鼠。应用体外培养的方法,加入不同浓度的TGF-β1,通过MTT法观察其对ASMC增殖的影响。实验分4组:对照组犤20mL/L胎牛血清(FCS)/低糖Dulbecco最低必须培养液(DMEM)犦,100mL/LFCS/DMEM组,10μg/LTGF-β1组和100μg/LTGF-β1组。结果:细胞培养第2天,MTT法检测的10μg/LTGF-β1组A值(0.36±0.043)和100μg/LTGF-β1组(0.37±0.050)明显高于对照组(0.126±0.052)(t=5.44,7.63,P<0.05)和100mLFCS/DMEM组(0.182±0.051)(t=5.97,5.88,P<0.05)。100mLFCS/DMEM组(0.38±0.034)于细胞培养第3天A值与对照组(0.20±0.035)比较增高(t=8.18,P<0.05)。镜下观察发现,10μg/LTGF-β1组和100μg/LTGF-β1组ASMC于培养第3天铺满96孔培养板,而100mL/LFCS/DMEM组细胞于第5天铺满培养板。结论:TGF-β1对气道平滑肌细胞的增殖具有明显的促进作用。 AIM:To observe the effects of transforming growth factor-β1 (TGF-β1)on p roliferation of airway smooth muscle cells (ASMCs) in rat. METHODS:ASMCs, obtained from a healthy male SD rats, were cultured in vitro in different concentration of TGF-β1 and its effect on proliferation of ASMCs wa s detected with MTT method. The ASMCs were divided into 4 groups: control group [2 mL/L fetal calf serum (FCS)/ Dulbecco minimum essential medium (DMEM)], 100 m L/L FCS/DMEM group,10 μg/L TGF-β1 group and 100 μg/L TGF-β1 group. RESULTS:On the 2nd day of cell culture, the A value detected through MTT in th e 10 μg/L TGF-β1 group(0.36±0.043) and 100 μg/L TGF-β1 group(0.37±0.050) were significantly higher than that in the control group (0.126±0.052)(t=5.44, 7.63,P< 0.05) and 100 mL/L FCS/DMEM group (0.182±0.051) (t=5.97,5.88,P< 0.05). On the 3rd day,the A value in the 100 mL/L FCS/DMEM group (0.38±0.034) were hig her than that in the control group (0.20±0.035) (t=8.18,P< 0.05).Under the micr oscope, the ASMCs in the 10 μg/L TGF-β1 group and 100 μg/L TGF-β1 group we re confluent in 96-well plateson the 3rd day, but 5th day in the 100 mL/L FCS/D MEM group.
