Akt在肌萎缩性脊髓侧索硬化症转基因鼠脊髓中的表达变化及意义 被引量:4

摘要 目的:探讨Akt在肌萎缩性脊髓侧索硬化症(ALS)发病中的作用,为临床治疗ALS提供实验依据.方法:选择ALS转基因鼠和野生型鼠各36只,分别在发病早期(95 d)、中期(108 d)和晚期(122 d)处死动物、分离脊髓,应用RT-PCR和免疫印迹检测Akt mRNA和蛋白水平的表达变化,应用免疫组织化学检测Akt阳性细胞的分布特点和表达规律.结果:ALS转基因鼠与野生型鼠相比,发病早期、中期和晚期脊髓组织中Akt mRNA和总蛋白均无明显变化,p-Akt(Thr308)蛋白表达均上调,发病中期和晚期p-Akt(Ser473)蛋白表达均上调.免疫组织化学显色结果显示,ALS转基因鼠和野生型鼠中p-Akt(Ser473)免疫阳性细胞主要分布在脊髓灰质前角.免疫荧光双标结果显示,Akt阳性细胞主要为神经元,少数为星形胶质细胞.结论:ALS转基因鼠脊髓组织中磷酸化Akt表达上调,Akt激活与ALS发病密切相关.
