摘要
线粒体甘油-3-磷酸酰基转移酶(mitochondrial glycerol-3-phosphate acyltransferase,GPAM)基因的主要作用是通过催化三酰基甘油和磷脂生物合成过程,从而促进动物机体甘油三酯(TAG)的生成,并通过中心和外周调节作用,对哺乳动物脂肪沉积,能量消耗,胴体质量以及整体的代谢进行调控。为揭示GPAM基因外显子多态性与西门塔尔牛胴体组成和肉质性状的相关性,本研究采用DNA池测序及PCR-RFLP的方法检测了131头34月龄中国西门塔牛群体该基因外显子中SNP位点的多态性,并与西门塔尔牛胴体组成、肉质性状及脂肪酸组成进行了关联分析。结果表明,在GPAM基因外显子区第20外显子2 823bp处检测到1个突变位点C>T,该SNP与中国西门塔牛的胴体组成性状(毛质量、胴体质量、净肉率、后部排酸后酸度、胴体长、胴体深、胴体胸深、牛鞭质量、头质量、前蹄质量、心脏质量)显著相关(P<0.05),与脂肪酸组成性状(花生酸、二十碳一烯酸)显著相关(P<0.05),但对肌间脂肪沉积无影响。本研究结果可为中国西门塔尔牛分子选育、进一步提高肉用性能提供依据。
The main function of mitochondrial glycerol-3-phosphate acyltransferase(GPAM)gene promotes the animals body triglyceride production through catalytic triacylglycero phospholipid biosynthesis,thus and regulates the energy consumption,carcass weight and fat deposition for mammals,the overall metabolism through the center and peripheral regulation.In this study,GPAM gene exon polymorphisms were detected using PCR-RFLP method in Chinese Simmental cattle population,and a relationship between GPAM gene SNP and cattle carcass composition and meat quality traits was analyzed.The results showed that a mutation 2 823 bp C>T in GPAM gene exon was detected,and showed a significant association with carcass traits(carcass weight,net meat rate,carcass length,carcass depth,heart weight,etc.)and fatty acid composition(arachidic acid)in Chinese Simmental cattle population(P<0.05).Our findings suggest that polymorphisms in GPAM gene exon might be one of the important genetic factors that influence carcass yield in beef cattle,and it may be a useful marker for carcass traits in future marker-assisted selection programmes in beef cattle breeding and production.
