肺炎支原体脂质膜蛋白诱导人支气管上皮细胞SPLUNC1的表达 被引量:3

摘要 [目的]观察不同浓度肺炎支原体脂质相关膜蛋白(MP-LAMPs)对人支气管上皮细胞(HBEC)表达短的上腭、肺及鼻咽上皮克隆1(SPLUNC1)的影响,探讨SPLUNC1在呼吸道感染中的作用及肺炎支原体(MP)感染与SPLUNC1表达的量效关系及时效关系.[方法]通过实时荧光定量PCR(FQ-PCR)检测不同浓度的MP-LAMPs刺激HBEC后,在不同时间点(2h、4h、8h、16h及24 h)SPLUNC1基因表达的变化;同时用Western-blot检测0.5 μg/mL MP-LAMPs刺激HBEC后不同时间点SPLUNC1蛋白表达情况.[结果]正常人HBEC有SPLUNC1表达;在相同时间点,不同浓度的MP-LAMPs均能诱导SPLUNC1基因表达上调,其中0.5 μg/mL s浓度组表达量最高,不同浓度组间比较差异有统计学意义(P<0.01);在同一浓度组,MP-LAMPs刺激细胞2h后SPLUNC1基因及蛋白表达量开始上调,并在4h时达高峰,随着时间延长逐渐下降,不同时间点组间比较差异有统计学意义(P<0.01).[结论]MP-LAMPs能诱导HBEC的SPLUNC1基因表达上调,并成一定的剂量依耐性,其表达量最佳浓度和时间点为0.5 μg/mL MP-LAMPs刺激HBEC 4 h.
