糖尿病模型大鼠白内障形成过程及枸杞多糖干预的实验研究 被引量:4

An Experimental Study on Formation Process of Cataract and Lycium Barbarum Polysaccharide Intervention in Diabetic Rats

摘要 目的观察STZ诱导大鼠糖尿病性白内障模型的建立,并观察中药枸杞提取物枸杞多糖(lycium barba-rum polysaccharide,LBP)对SD大鼠糖尿病性白内障形成的干预作用。方法将40只大鼠称取体质量后按随机数字表法随机分为空白组、模型组、枸杞多糖组。采用一次性腹腔内注射链脲佐菌素(STZ)(65mg·kg^(-1))的方法建立大鼠糖尿病模型,枸杞多糖组大鼠自STZ注射2周后每日给予枸杞多糖200mg·kg^(-1)灌胃,共灌14周。STZ注射后72h、1周及注射后每2周监测大鼠饮食、体质量及血糖的变化,并在裂隙灯下观察大鼠晶状体混浊的进展,进行分期并记录各时间点大鼠的例数。结果将第1、8、16周各组大鼠的晶状体形态分期后各期大鼠的例数应用秩和检验进行对比发现:与模型组相比,枸杞多糖组大鼠晶状体混浊程度发展较为缓慢(P<0.05)。结论单次大剂量STZ诱导的糖尿病模型大鼠白内障发病缓慢,较接近人糖尿病性白内障的特点;中药枸杞提取物LBP对SD大鼠糖尿病性白内障的形成具有一定的延缓作用。 Objective To observe diabetic cataract in model rats induced by STZ,and to investigate the effect of the Lycium barbarum polysaccharides(LBP) extract on formation of diabetic cataract in SD - rats.Methods 40 rats which had been weighted were randomly divided into control group,model group,and Lycium barbarum polysaccharides group(200mg·kg<sup>-1</sup>).Using a single intraperitoneal injection with streptozotocin (STZ)(65mg·kg<sup>-1</sup> ) to establish the model of diabetic rats,after STZ injection for 2 weeks,Lycium barbarum polysaccharide 200mg·kg<sup>-1</sup> were administered to Lycium barbarum polysaccharides group daily,a total of 14 weeks.After STZ injection for 72h,1 week,2 weeks,the changes of diet,body weight and blood glucose were monitored and the progression of lens opacity was observed at the slit lamp.Results At the first week,eighth week,and sixteenth week,len shape stage of the rats and the quality showed by rank sum test: compared with the model group,Lens opacity in Lycium barbarum polysaccharides group developed more slowly significantly(P &lt; 0.05).Conclusion Single large dose STZ induced cataract in diabetic rats develop slowly,which is similar to the features of human diabetic cataract;Extract of LBP has certain delayed effect on the formation of diabetic cataract in SD - rats.
