rhBNP对心衰家兔模型心脏保护机制的研究 被引量:1

摘要 目的探讨重组人脑钠肽(rhBNP)对心衰家兔模型心脏的保护作用及可能机制。方法以阿霉素诱导心衰家兔模型,同时设对照组、模型组和治疗组。连续治疗4周后,腹主动脉采血行酶联免疫吸附测定法行血管紧张素Ⅱ检测;HE染色观察心脏组织病理变化;RT-PCR检测心肌Ⅰ、Ⅲ型胶原的mRNA表达情况。结果实验结果表明心衰家兔心肌中Ⅰ、Ⅲ型胶原含量呈高表达,且心肌组织结构发生明显的病理改变。rhBNP在一定程度上能拮抗血管紧张素II(AngII)的活性,对HF的治疗有一定的疗效。结论模型组心肌Ⅰ、Ⅲ型胶原含量明显高于正常对照组,具有显著差异性(P<0.05);与模型组比较,rhBNP治疗组心肌Ⅰ、Ⅲ型胶原含量的表达下降(P<0.05)。 Objective To investigate the protection function and the possible mechanism of rhBNP to cardiac of heart failure rabbit model.Methods Using adriamycin induced heart failure rabbit model,and establishing the control group,model group and the treatment group.After 4 weeks continuous treatment,using abdominal aortic blood for determination angiotensin Ⅱ,Enzyme linked immunosorbent assay method,pathological section and HE staining for detecting cardiac tissue pathological changes,and RT-PCR detection for the mRNA expression of colⅠand col Ⅲ.Results The experimental results show that colⅠand col Ⅲ had a high expression in heart failure rabbits,and myocardial structure has obvious pathological changes.RhBNP can protect against the activity of angiotensin II(AngII) to a certain extent.So the treatment of BNP to HF has a certain curative effect.Conclusion The mRNA expression of colⅠand col Ⅲ of model group is higher than control group,and it has a significant difference(P<0.05).Compared with model group,the expression of col Ⅰand col Ⅲ of BNP treatment group had a decline(P<0.05).
