β_2受体阻断剂ICI 118551预防大鼠主动脉内注射小剂量肾上腺素所致的血压下降 被引量:2

摘要 目的肾上腺素浸润注射或气管内应用可引起血压明显的下降,其确切机制尚不清楚。文中观察大鼠主动脉内注射小剂量肾上腺素降低血压作用,以及静脉预先注射特异性β2受体阻断剂ICI 118551对预防此类血压下降的作用。方法成年雄性SD大鼠48只随机均分为4组(n=12):等渗盐水对照组(S组)、ICI 118551+等渗盐水组(I组)、等渗盐水+肾上腺素组(E组)及ICI 118551+肾上腺素组(IE组)。S组和I组先分别静脉注射等渗盐水0.2ml和ICI 118551 0.5mg/kg,20 min后在大鼠主动脉处注射等渗盐水0.2 ml;E组和IE组先分别静脉注射等渗盐水0.2 ml和ICI 118551 0.5 mg/kg,20 min后在大鼠主动脉处注射肾上腺素0.1μg/kg。记录注射前基础值(T0)和注射后5、10、15、30s及1、2、3、4、5min(T1-9)时的平均动脉压(mean arterial pressure,MAP)和心率(heart rate,HR)。结果与T0时相比,E组于T2-4时MAP下降(P<0.05),并于T5时恢复至基础值水平;其余3组各时间点MAP未见明显改变。4组HR各时间点未见明显改变。结论主动脉内注射0.1μg/kg肾上腺素可引起大鼠血压短暂下降,静脉预先注射ICI 118551可预防此类血压的下降。 Objective Infiltrated injection or endotracheal administration of epinephrine can decrease blood pressure,but its mechanism has not been well investigated.This study was to determine whether intra-aortic injection of epinephrine can decrease blood pressure,and whether pretreatment with ICI 118551 can prevent this fall in rats.Methods Forty-eight male SD rats were equally randomized to 4 groups: saline control(S),ICI 118551+saline(I),saline+epinephrine(E),and ICI 118551+epinephrine(IE).The S and I groups were intravenously injected with 0.2 ml saline and ICI 118551 at 0.5 mg/kg,respectively,20 min before intra-aortic injection of 0.2 ml saline,while the E and IE groups received the same intravenous medication 20 min before intra-aortic injection of 0.2 ml epinephrine at 0.1 μg/kg.Mean arterial pressure(MAP) and heart rate(HR) were recorded before intra-aortic injection(T0) and 5s,10s,15s,30s,1 min,2 min,3 min,4 min,and 5 min(T1-9) after it.Results Compared with T0,MAP decreased at T2-4 and returned to the baseline at T5 in the E group(P<0.05),but had no significant changes in the other 3 groups at all time points.None of the 4 groups showed remarkable changes in HR at any time.Conclusion Intra-aortic injection of epinephrine at 0.1 μg/kg can transitorily decrease blood pressure in rats,and pretreatment with ICI 118551 at 0.5 mg/kg can prevent this fall.
