摘要
对脓毒症犬模型进行体外循环血液净化治疗,观察高容量血液滤过联合内毒素吸附对其内皮细胞的影响。采用高容量血液滤过联合内毒素吸附治疗。三组动物分别在注射内毒素前(T1)、注射后60 min(T2)、120 min(T3)、240 min(T4)、480 min(T5)时采取血液标本,检测血清内皮素(Endothelin,ET)、血管性假血友病因子(von willebrand factor,vWF)、内毒素(Lipopolysaccharide,LPS)水平。除对照组外,四组动物造模后血浆ET、vWF和LPS浓度均较造模前升高5~6倍,治疗后4 h,HVHF组ET、vWF和LPS浓度缓慢下降,而序贯组在治疗后2 h,即可见ETv、WF和LPS浓度同步下降,与其他两组相比有显著性差异。高容量血液滤过联合内毒素吸附治疗可降低血浆LPS、ET和vWF水平,调控炎症反应,保护内皮细胞功能,防治DIC,从而在阻断脓毒症、MODS向多脏器功能衰竭(Multiple organ failure,MOF)的治疗中发挥了重要的作用。
Using high volume hemofiltration(HVHF)to treat animal model,to evaluate the influences of hemoperfusion on sepsis.Blood samples were collected from all animals before endotoxin infusion(T1,baseline) and at 60(T2),120(T3),240(T4),480 min(T5) after endotoxin infusion.The dynamic change of ET,vWF.In addition to the control group,the concentrations of ET,vWF and LPS of four group animals were increased significantly.After four hours of treatment,the concentrations of LPS and cytokines in peripheral blood decreased significantly after HF.In the treatment on sepsis,HF can regulate inflammatory response effectively 、decrease the concentrations of LPS,ET and vWF,HF can attenuate the circulating level of cytokines,improve major organ function;protect the function of endothelial cell.
