摘要
目的探讨去甲斑蝥素对人肝癌HepG2细胞系抑制和诱导凋亡作用。方法采用MTT法检测细胞的生长活性,应用流式细胞仪检测、透射电镜下观察去甲斑素处理HepG2细胞后细胞周期阻滞和细胞凋亡的发生情况。结果 5~40μg/ml去甲斑蝥素均能抑制HepG2细胞增殖(P<0.05),其抑制率与作用时间及NCTD浓度相关。流式细胞仪检测发现,去甲斑蝥素处理HepG2细胞后,实验组G2/M细胞明显多于对照组,P<0.05,提示细胞周期停滞于G2/M期。透射电镜下可见,HepG2细胞出现凋亡形态学改变。结论去甲斑蝥素对HepG2细胞有抑制作用,可能与阻滞细胞周期及诱导细胞凋亡有关。
Objective To study the inhibitory effect of Norcantharidin(NCTD)on human Hepatoma HepG2 cells.Methods MTT assay was used to determine the cell growth inhibitory rate in vitro,flow cytometry and electron microscopes were used to study the effect of the changes of cell cycle and the apoptosis of HepG2 cells before and after NCTD treatment.Results MTT showed that 5~40 μg/ml NCTD inhibited HepG2 cells growth.NCTDinhibited HepG2 cell growth in a time-and dose-dependent manner.The result of flow cytometry showed that cells in G2/M phase were increased after exposure to NCTD.Morphological apoptosis of HepG2 cells could be seen under electron microscope.Conclusion NCTD inhibits HepG2 cells proliferation and induces HepG2cells apoptosis
