摘要
目的研究碱性成纤维生长因子(bFGF)和表皮生长因子(EGF)对胚胎神经干细胞生长和分化的影响。方法从孕11~13d大鼠胚胎神经管取神经干细胞,原代体外培养中加入bFGF和EGF,观察对神经干细胞胞体发育及突起生长的影响。结果EGF主要影响神经干细胞的增殖,也可以促进细胞突起的生长,但胞体直径与相应的对照组无显著差异;bFGF可促进胞体发育及突起的生长;两种因子联合应用,生长作用优于单独应用组。结论EGF和bFGF可以促进原代培养胚胎神经干细胞的生长和分化。
Objective To study the effects of bFGF and EGF on the embryonic neural stem cells in primary culture.Method Embryonic E12 rats were abtained from anesthetized timed pregnant Sprague-Dawley rats. Neural stem cells were dissociated from embryonic neural tube.The bFGF and EGF were added either alone or in combination into the culture medium.Results EGF mainly affected the proliferation of neural stem cells and slightly promoted the outgrowth of the processes, but could not stimulated the development of the cell bodies; while bFGF could stimulated the development of the cell bodies and processes.The data of EGF+bFGF group were superior to that of bFGF or EGFgroup. Conclusion bFGF and EGF both can enhance the outgrowth and differentiation of neural stem cells in primary culture.
