摘要
目的探讨大鼠坐骨神经切断后胶质细胞源性神经营养因子GDNFmRNA在远侧断端的表达变化及其意义。方法切断SD大鼠左侧坐骨神经,不同时间、半定量RT-PCR方法,观察远侧断端GDNFmRNA的表达变化。结果GDNFmRNA在坐骨神经切断前微量表达,坐骨神经切断后远断端表达逐渐增加(1、7、14、28d分别增加20%、60%、85%、90%)。结论坐骨神经切断导致“胞体-轴突-靶器官”轴中断,使靶组织产生GDNF聚集于坐骨神经远断端,表现为GDNFmRNA高表达。
Objective To investigate the gene expression o f GDNFmRNA on distant side nerve afte r sciatic nerve was cut in rats.Method The specimen of sciatic nerve of distant side was taken respectively befo re and after sciatic nerve severed.T he level of GDNFmRNA on distant side was observed and comp ared before and after sciatic nerve c ut,using semi -quantitative RT -PCR method withβ-actin as a inner consult.Result Much minim expression of GDNFmRNA wa s found on sciatic nerve before opera tion,but it increased 20%of the ex-pression quantity 24hours after sciatic nerve cut,60%7d later,85%14d a nd 90%28d respectively on distant br eaking side.Conclusion The enhancing of GDNFmRNA level on di stant sciatic nerve breaking side sh ould be caused by the gathering of GDN F which was produced by target tissue after intermitting ofneuron -axon -targetaxis when sciatic nerve cut.This pro vided one of theory foundations for f oreign GDNF to be used in treating SCI.
