摘要
目的完善大鼠肝星形细胞(HSC)的体外分离及培养方法,为肝纤维化的基础实验研究提供技术支持。方法选用Wistar大鼠经消化酶灌注肝脏后,用分离液分离HSC,通过观察其自发荧光及其显著特征性结构的脂肪染色、细胞免疫化学染色等方法鉴定。结果分离得到HSC,细胞得率为2×107/只,存活率为96%。结论通过本法成功分离及培养HSC,且方法经济、简便、稳定,有利于原代大鼠肝星状细胞的生物学研究。
Objective To improve methods of extracorporeal isolation and culture of rat hepatic stellate cells.Methods HSC was islated with isolation liquid after the Wistar rats were given liver perfusion,methods of fat stains and cellular immunochemistry stains were identified through observation of its spontaneous fluorescence and significant features.Results The yield of HSC got by this method was about 2×107 per rat liver,purity and activity of cells were 96%.Conclusion This work providee a good and economic metho...
