摘要
从感染致弱的Ⅰ型马立克氏病病毒(MDV)814株的细胞中提取病毒基因组DNA,以digoxingenin标记MDV强毒GA株糖蛋白B(gB)基因片段作为探针,进行Southern-blot杂交定位。将包含MDV 814株gB基因的基因组片段克隆于pUC19质粒中。酶切鉴定后,进行序列分析。结果表明该 gB基因具有高度的保守性,编码区序列同已发表的Ⅰ型马立克氏病病毒 gB基因序列完全一致,但启动子上游区域有差异。
The genomic DNAs of the strain 814 of Marek s disease virus(MDV) were extracted from chickes embryo fibroblasts infected by the strain 814 of MDV. The gene encoding glycoprotein B (gB) of the strain 814 of MDV was oriented by southern-blot. Futhermore, the gB was cloned into plasmid pUC19, and the entire nucleotide sequence of the gene was determined. The results indicated that gB of MDV type Ⅰ was highy conservative, and the encoding sequence of gB of strain 814 was the same as that of strain RBIB of MDV, ...
