摘要
目的观察吡格列酮对缺血再灌注损伤心肌细胞的保护作用,为吡格列酮治疗心肌缺血再灌注损伤提供理论基础和实验依据。方法体外培养Wistar大鼠乳鼠心室肌细胞,建立心肌细胞缺氧复氧模型。比色法检测各组培养基中SOD活力和MDA的含量;采用碘化丙啶标记流式细胞术和Hochest33258免疫荧光法检测心肌细胞凋亡率。结果吡格列酮使培养的缺血再灌注损伤心肌细胞培养基中的MDA含量降低;SOD活力升高(P<0.05);凋亡指数显著降低。结论吡格列酮减轻离体缺血再灌注损伤心肌细胞凋亡,具有保护作用。
