摘要
为构建转基因SD大鼠,用3种不同转基因构件p279-D5、p279-D5B、p279-SV40Tag对SD大鼠受精卵进行显微注射和胚胎移植试验,移植出生仔鼠经PCR检测筛选首建鼠。结果表明:以每只20 U的孕马血清促性腺激素和人绒毛膜促性腺激素的剂量组合对5周龄组大鼠达到了最佳超数排卵效果。3种构件经PCR检测后都获得了阳性后代,其外源基因整合率分别为p279-D51.4%(5/350)、p279-D5B 1.1%(3/274)和p279-SV40Tag 1.3%(3/230)。
For conceiving sprague-dawley(SD) transgenic rat,three different DNA-constructs which is p279-D5, p279-D5B,and p279-SV40Tag into rat zygote and transferred survival embryos into pseudopregnant were microinjected. The offsprings of pregnancies were detected by PCR to screen founder rats.The following results were obtained.The most effective supervoulation was 5 weeks SD female rats injected with 20 U PMSG and HCG each female rat.Five founder transgenic rats of p279-D5 from 350 offsprings of pregnancies(1.4%)...
