摘要
目的:探讨驱虫斑鸠菊对人 A375黑素瘤细胞株分泌酪氨酸酶活性、黑素合成以及细胞内酪氨酸酶基因表达水平的影响。方法:采用酶学方法研究驱虫斑鸠菊对酪氨酸酶活性的影响;采用475 nm 比色法测定黑素含量;采用 TRIzol 试剂盒提取总 RNA,再用半定量逆转录聚合酶链反应扩增 cDNA,检测酪氨酸酶基因表达水平。结果:驱虫斑鸠菊可以增强 A375人黑素瘤细胞酪氨酸酶活性和黑色素合成能力,还可增强酪氨酸酶基因表达。结论:驱虫斑鸠菊可能从基因水平增强酪氨酸酶活性,促进黑素合成。
Objective:To study the effect of Vemonia Anthelmintica(VA)on tyrosinase secretion of human melanoma cell strain A375,its ac- tivity,the melanin synthesis and intracellular tyrosinase gene expression.Methods:Tyrosinase activity was measured by zymologic method.The melanin was detected by colorimetry with absorption at 475nm.Total RNA was extracted using Trizol kit and cDNA was augmented with semi-quantitative reverse transcription polymerase chain reaction(RT-PCR)to test the gene expression.Results:VA could ...
