摘要
目的:探讨首茸方缓解长期使用左旋多巴(L-DOPA)治疗帕金森病(PD)患者的疗效减退的机理。方法:应用脑內微透析技术、水杨酸捕获活体内羟自由基和高效液相-电化学检测(HPLC-ED)法观察首茸方对 L-DOPA 诱发清醒的、由6-羟基多巴胺所致的 PD 大鼠脑纹状体细胞外液中羟自由基增高的影响。结果:首茸方给药7天显著增加 PD 大鼠纹状体细胞外多巴胺(DA)的水平,并降低 PD 大鼠脑內羟自由基水平;L-DOPA 处理可以增加脑内羟自由基的水平,PD 大鼠纹状体细胞外2,5-DHBA 水平明显升高,首茸方可降低其增高。结论:左旋多巴治疗 PD 疗效衰减的机制可能部分原因是由于氧自由基持续增高损害了残留的 DA神经元。本研究中首茸方不仅能增加脑内 DA 的水平,还能抑制 L-DOPA 引起的羟自由基的增加,可认为是一种中药的神经元保护剂。
