摘要
目的研究抗血管内皮生长因子(VEGF)单克隆抗体对肝缺血再灌注损伤的作用及其机制。方法雄性SD大鼠30只,随机分为A组(抗VEGF单克隆抗体干预组)、B组(缺血再灌注组)、C组(手术对照组),其中A、B组建立肝缺血再灌注损伤模型。A组于缺血前15min经腹腔注射抗VEGF单克隆抗体(100μg/300 g);B组于缺血前15 min经腹腔注射等量生理盐水;C组仅作麻醉、开腹、不阻断血流。术后6h,分别测定血肝功能酶(ALT、AST、LDH)、肝脏VEGF表达、肝组织病理改变、肝细胞凋亡情况,肝组织髓过氧化物酶(MPO)的含量。结果A组血ALT、AST、LDH及肝组织MPO显著低于B组(P<0.01),肝脏局部VEGF的表达明显减少,肝组织形态学无明显改变,细胞凋亡显著减少。结论抗VEGF单克隆抗体能有效减轻大鼠肝脏缺血再灌注损伤,缓解肝功能下降。
