抗ABCA1自身抗体在SLE患者动脉粥样硬化发病机制中的意义 被引量:3

Detection of the auto-antibodies against ATP-binding cassette transporter A1 and its significance in the pathogenesis of atherosclerosis in patients with systemic lupus erythematosis

摘要 检测系统性红斑狼疮(SLE)患者血清中抗三磷酸腺苷结合盒转运子(ABCA1)抗体及探讨其致SLE早发动脉粥样硬化(AS)的机制。采用免疫印迹法检测75份SLE患者血清中抗ABCA1自身抗体。75份性别年龄匹配正常人血清为对照。观察抗ABCA1抗体阳性患者血清IgG对细胞胆固醇流出的影响。结果显示,SLE患者血清中存在抗ABCA1抗体,阳性率为29.3%,正常人中无一阳性(P<0.05)。SLE有斑块组的抗ABCA1抗体阳性率高于SLE无斑块组(43.8%和16.3%,P<0.05)。纯化的抗ABCA1抗体阳性SLE患者IgG体外可抑制THP-1细胞内胆固醇的流出,抑制率最高可达28.7%,正常人IgG抑制率相比有显著性差异。实验表明,SLE患者血清中抗ABCA1抗体阳性率明显高于正常人,且纯化的抗AB-CA1抗体阳性患者IgG在体外能抑制细胞内胆固醇的流出,从而促进SLE患者早发动脉粥样硬化的发生。 To detect the auto-antibodies against ATP-binding cassette transporter A1(ABCA1)in serum samples of patients with systemic lupus erythematosis(SLE) and to explore its significance in the pathogenesis of atherosclerosis in these patients,the auto-antibodies against ABCA1 in sera of 75 SLE patients and 38 healthy individuals were detected by immunoblotting assay,and the effect of the anti-ABCA1 antibody IgG on the efflux of cholesterol from THP-1 cells was examined.
