摘要
目的研究核酸对铅中毒大毒淋巴细胞、肝细胞和脑细胞DNA损伤的保护作用。方法纯种雄性健康Wistar大鼠,随机分为4组:空白组、模型组、核酸低剂量(200 mg/kg体重)组、核酸高剂量(700 mg/kg体重)组。空白组正常饮水,其余3组饮0.8%醋酸铅水溶液;核酸组灌胃核酸,空白组、模型组灌蒸馏水5 w后,采用单细胞凝胶电泳技术对淋巴细胞、肝细胞和脑细胞DNA损伤进行分析。结果模型组大鼠淋巴细胞、肝、脑细胞DNA的迁移率(50.25±6.21、56.25±6.71、38.13±5.28)和尾长(23.25±4.23、27.63±4.98、37.63±5.37)显著高于空白组(6.88±1.13、8.25±1.28、8.63±1.19;6.75±1.17、7.88±0.99、5.13±0.84),核酸组大鼠淋巴细胞、肝、脑细胞DNA的迁移率(低剂量组:41.25±6.96、42.25±6.09、28.13±3.76,高剂量组:35.38±5.93、39.25±7.17、23.88±3.23)和尾长(低剂量:16.50±4.24、19.25±3.54、26.00±5.24,高剂量:13.63±2.62、18.50±5.48、22.38±5.61)显著低于模型组,差异均具有统计学意义(均P<0.05)。结论慢性铅染毒可导致大鼠淋巴细胞、肝、脑细胞DNA损伤,饮食核酸对淋巴细胞、肝、脑细胞DNA损伤具有一定的保护作用,可减轻铅中毒引起的氧化损伤。
Objective To observe the protective effect of nucleic acid on DNA damage of peripheral blood lymphocytes,liver cells,brain cells,in lead-poisoning rats.Methods Male rats were divided into four groups which were the blank group,model group,nucleic acid low-dose group,nucleic acid high-dose group. The bland group normally drank water,and other three groups drank 0.8% lead acetate solution.The nucleic acid groups were poured for distilled water.After 5 weeks,the DNA damage of peripheral blood lymphocytes,liver cells and brain cells from the rats were analyzed with single cell gel electrophoresis.Results The mobility(50.25±6.21,56.25±6.71,38.13±5.28)and tail length(23.25±4.23,27.63±4.98,37.63±5.37)of peripheral blood lymphocytes,liver cells and brain cells DNA of the model group were significantly higher than those of blank group(6.88±1.13,6.75±1.17).The mobility and tail length peripheral blood lymphocytes, liver cells and brain cells DNA of nucleic acid groups were significantly lower than those of model group(P<0.05).Conclusion Chronic lead-poisoning causes DNA damage of the peripheral blood lymphocytes,liver cells and brain cells.Dietary nucleic acid could protect DNA damage of peripheral blood lymphocytes,liver cells and brain cells in rats and reduce the oxidative damage from lead-poisoning.
