Optimization of Extraction Process of Total Flavonoids from Lysimachia Christinae Hance by Orthogonal Design

摘要 Objective: to study the alcohol extraction process of total flavonoids from Lysimachia christinae Hance. Methods: ultraviolet spectrophotometry was used to determine the absorbance and calculate the extraction rate of total flavonoids from Lysimachia christinae Hance. Single factor experiment and orthogonal experiment design were adopted respectively. The factors of investigation were ethanol concentration, solid-liquid ratio, extraction time and extraction times. Each factor was set at 3 levels, with ethanol concentration of 30%, 50% and 70%. Material-liquid ratio: 1:5, 1:10, 1: 15;Extraction times: 1, 2 and 3 times;Extraction time: 1h, 1.5h, 2h, the optimal extraction process of total flavonoids from Lysimachia christinae was optimized. Results: the method has good precision, stability and reproducibility. The sample addition recovery rate meets relevant standards;The standard curve has good linearity, and the regression equation is: y = 1.1138x+0.046 (R2 = 0.999);The optimal extraction process of total flavonoids from Lysimachia christinae Hance is: extract with 10 times of 50% ethanol for 3 times, 1.5 hours each time. Under these conditions, the average extraction rate of total flavonoids from Lysimachia christinae was 2.98%. Conclusion: the optimized extraction process has simple operation and good stability, which can provide a theoretical basis for further research and development of the traditional Chinese medicine Lysimachia christinae.
