摘要
目的探讨lncRNA SNHG1(核仁小分子RNA宿主基因1)对乳腺癌细胞侵袭及转移能力的影响,并探索潜在机制。方法利用划痕实验和Transwell实验检测SNHG1和miR-361-3p对乳腺癌细胞迁移和侵袭能力的影响,利用双荧光素酶报告基因实验检测SNHG1是否可结合miR-361-3p,miR-361-3p是否能够结合基质金属蛋白酶1(MMP-1),利用实时荧光定量PCR和蛋白免疫印记实验检测SNHG1与miR-361-3p以及MMP-1表达的关系。结果抑制SNHG1和增加miR-361-3p的表达后,细胞迁移和侵袭能力均降低(P<0.01)。双荧光素酶报告基因实验结果显示SNHG1能够吸附miR-361-3p(P<0.01),miR-361-3p可结合MMP-1(P<0.01)。PCR结果显示SNHG1低表达使得miR-361-3p的表达增加(P<0.01),蛋白免疫印记实验结果显示抑制SNHG1的表达可降低MMP-1的表达(P<0.01)。结论SNHG1/mir-361-3p/MMP-1轴调控乳腺癌细胞迁移和侵袭。
Objective To investigate the effects of LncRNA SNHG1 on the invasion and metastasis of breast cancer cells and explore the underlying mechanisms.Methods Wound healing assays and Transwell assay were used to detect the effect of SNHG1 and miR-361-3 p on the migration and invasion of breast cancer cells.The dual luciferase reporter gene assay was used to detect whether SNHG1 can bind to miR-361-3 p and whether miR-361-3 p can bind to MMP-1.Real-time PCR and Western blot were performed to detect the relationship between SNHG1 and miR-361-3 p,and MMP-1.Results After inhibiting the expression of SNHG1 and promoting the expression of miR-361-3 p,cell migration and invasion ability were decreased(P<0.01).Dual luciferase reporter gene assay showed that SNHG1 could directly bind with miR-361-3 p(P<0.01),and miR-361-3 p can bind to MMP-1.Real-time PCR revealed that knockdown SNHG1 could promote the expression of miR-361-3 p(P<0.01).Western blot showed that inhibition of SNHG1 reduces MMP-1(P<0.01).Conclusion The SNHG1/mir-361-3 p/MMP-1 axis regulates breast cancer cell migration and invasion.
