B细胞CD23的表达与变应性鼻炎的相关性研究

摘要 目的:研究外周血及鼻黏膜中CD23的表达与变应性鼻炎的相关性,探讨变应性鼻炎治疗的新方法。方法:采用ELISA法、流式细胞术、HE染色和免疫荧光法分别检测变应性鼻炎组和对照组血清中总IgE、特异性-IgE、IFN-γ、IL-10、IL-4、CD19^(+)CD23^(+)B细胞、CD19^(+)CD5^(+)CD1d^(+)B细胞、CD3^(+)CD4^(+)T细胞数量、百分率以及B细胞CD23表面密度,鼻腔黏膜上CD23、CD19、IL-10的免疫荧光表达。结果:与对照组比较,变应性鼻炎组CD23平均荧光强度明显增强,CD19及IL-10表达明显减弱;变应性鼻炎组血清中总IgE、特异性-IgE、IL-4水平明显高于对照组;B细胞CD23的表面密度与总IgE、IL-4、特异性-IgE呈正相关,与IL-10、IFN-γ呈负相关。结论:B细胞CD23的表达,可能通过依赖Bregs分泌IL-10介导的T细胞活化参与变应性鼻炎的发生发展,有望成为控制和治疗变应性鼻炎的新方法。 Objective:To study the correlation of CD23 expression in peripheral blood and nasal mucosa with allergic rhinitis,and to investigate the new treatment for patients with allergic rhinitis.Methods:Total IgE,IgE specificity,IFN-γ,IL-10,IL-4,CD19^(+)CD23^(+),B cells and CD19^(+)CD5^(+)CD1d^(+)B cells,the number and percentage of CD3^(+)CD4^(+)T cells,and surface density of CD23 on B cells,and the immunofluorescence expressions of CD23,CD19 and IL-10 in nasal mucosa in the allergic rhinitis group and control group were detected by the respective ElLSA method,flow cytomentry,HE staining and immunofluorescence.Results:Compared with the control group,the average fluorescence intensity of CD23 in the allergic rhinitis group was significantly enhanced,and the expressions of CD19 and IL-10 were significantly decreased;the total IgE,IgE specificity and IL-4 levels in the allergic rhinitis group were obviously higher than those in the normal control group;the surface density of CD23 on B cells was positively correlated with total IgE,IL-4 and IgE specificity,but was negatively correlated with IL-10 and IFN-γ.Conclusion:The expression of CD23 on B cells may be involved in the development of allergic rhinitis through IL-10-mediated T cell activation dependent on Bregs secretion,which is expected to be a new method to control and treat patients with allergic rhinitis.
