基于线粒体COI基因的中华鳖3个品系遗传多样性分析 被引量:3

Genetic Diversity of Three Pelodiscus sinensis Strains Based on COI Gene Sequence

摘要 中华鳖(Pelodiscus sinensis)是我国重要的养殖渔业生物。近年来,因其自然野生种群数量不断减少,地方优良种质资源日益受到重视。为了深入了解目前重要养殖中华鳖品系的遗传本底现状,以期为中华鳖的保护及地方优良种质资源的挖掘利用提供依据,本研究以中华鳖淮河品系、黄河品系和日本品系为研究对象,通过对93个个体的COI基因序列进行分析,结果发现其序列共有57个核苷酸变异位点,其中单位点碱基突变11个,简约信息位点46个。所有序列可定义为21种单倍型,单倍型多样性为0.858,核苷酸多样性为0.004,表明当前中华鳖的遗传多样性比较丰富。在3个品系中,日本鳖遗传多样性最高,淮河鳖次之,黄河鳖最低。群体间的遗传分化指数为0.4689~0.6481,基因流为0.1357~0.283,说明3个品系间存在较大的遗传分化。建议在今后的保护和利用中,应注重中华鳖的遗传多样性保持,合理选种选配,加强良种利用,以促进优质高效中华鳖产业的高质量发展。 Chinese soft-shelled turtle,Pelodiscus sinensis,is one of the important economical fishery species in China.In the past decades,the natural wild populations of P.sinensis have decreased sharply and local turtle resources have been payed more attention.In order to understand the genetic background and provide reference for protecting and exploiting local P.sinensis germplasm resources,93 individuals of Huaihe River,Yellow River and Japan strains were analyzed base on COI gene sequence.The results showed that 57 polymorphic sites were observed,which include 11 singleton variable sites and 46 parsimony information sites.21 haplotypes were defined in three strains of P.sinensis.The overall haplotype and nucleotide diversity were 0.858 and 0.004,respectively,which indicated abundant genetic diversity.Level of genetic diversity was the highest in Japan strain and the lowest in Yellow River strain.Fixation index(Fst)and gene flow(Nm)ranged from 0.4689 to 0.6481 and 0.1357to 0.283,respectively.Significant genetic differentiation was found among Huaihe River,Yellow River and Japan strains.Thus,some measures should take to maintain the genetic diversity,perform scientific selection and mating,and utilize excellent turtle germplasm resources to sustainable development of P.sinensis industry.
