摘要
DNA甲基化作为一种重要的表观遗传调控,在鱼类体色形成方面发挥重要作用。本研究通过对豹纹鳃棘鲈(Plectropomus leopardus)基因组范围内的DNA甲基化结合蛋白(methyl-CpG-binding domain,MBD)家族进行分析,探究其家族成员序列特征及其在豹纹鳃棘鲈体色形成中的作用。利用豹纹鳃棘鲈基因组数据,共鉴定到10个mbd基因,分别命名为mbd1~mbd10。理化性质分析发现MBD家族蛋白分子量介于26.55~288.08kD,理论等电点在5.16~9.78,均属于不稳定蛋白。DNA结构分析显示MBD家族基因的外显子和内含子数量及结构均不一样。豹纹鳃棘鲈MBD蛋白均含有该基因家族典型的MBD结构域,序列比对显示MBD结构域保守性相对较低,但MBD-a及MBD-c结构域序列高度保守。MBD家族进化树分析发现MBD8~MBD10、MBD1~MBD3序列保守分别聚为一支,MBD5~MBD7聚为一支但保守性较前两支较低。Motif分析结果显示,MBD1~MBD3、MBD4~MBD6、MBD8~MBD10保守性较高,与进化树分析结果一致。豹纹鳃棘鲈不同体色转录组数据分析发现,mbd2、mbd6及mbd9基因在不同体色转录组中差异显著,暗示其在豹纹鳃棘鲈体色形成中的重要作用。本研究为进一步研究豹纹鳃棘鲈体色形成表观遗传调控提供了基础资料。
DNA methylation is one of an epigenetic regulation,which plays an important role in the formation of fish body color pigments synthesis.In the present study,genomic DNA methyl-CpG-binding domain(MBD)family of Plectropomus leopardus was analyzed to explore its potential in the body color formation of P.leopardus.Based on the genome data of P.leopardus,a total of 10 mbd genes were identified,named mbd1~mbd10.The molecular weight of MBD family ranged from 26.55 to 288.08kD and the theoretical isoelectric point ranged from 5.16 to 9.78,which indicated that they belong to unstable proteins.DNA structure analysis showed that the number of exons and introns of MBD family genes are different.All the MBD proteins contained the typical MBD domain.Sequence alignment showed that the MBD domain was relatively less conserved,and the MBD-a and MBD-c domain sequences were highly conserved.The phylogenetic tree of MBD family showed that the sequences between MBD8~MBD10 and MBD1~MBD3 were conserved and clustered into one branch,respectively and MBD5~MBD7 clustered into one branch.The results of Motif analysis showed that MBD1~MBD3,MBD4~MBD6,and MBD8~MBD10 were highly conserved,which was consistent with the results of phylogenetic tree.Transcriptome analysis of different body color of P.leopardus showed that mbd2,mbd6 and mbd9 genes showed significantly different expression,suggesting their important role in the body color formation of P.leopardus.The present study provided fundamental materials for epigenetic regulation research of P.leopardus body color.
