利用SSR荧光标记构建七十二个莲属荷花品种指纹图谱 被引量:8

Establishment of Seventy-two Lotus (Nelumbo spp.) Cultivars Fingerprints Using SSR Fluorescent Markers

摘要 以72个荷花品种为试材,采用荧光标记SSR结合毛细管电泳的方法,研究了不同荷花品种的遗传多样性,并构建了指纹图谱,以期为荷花种质资源的收集和分类鉴定提供依据。结果表明:利用9对SSR荧光引物,通过毛细管电泳技术对72个荷花品种进行检测,共检测到51个多态位点,多态位点数平均为5.67;多态性信息含量PIC值变化范围为0.34~0.76,平均值为0.56。利用其中6对引物CL04、SSR449、SSR412、CL01、CL16和CL10可区分72份供试品种。利用引物扩增带型组合法,构建了72个荷花品种的指纹图谱并进行聚类分析,在遗传相似系数0.535处供试荷花材料可分为三大类群。该研究结果可为荷花种质资源鉴定和分子育种提供参考依据。 Seventy-two lotus varieties were used as materials,the genetic diversity of different varieties was studied by fluorescence labeled SSR and capillary electrophoresis,and the fingerprints were constructed in order to provide basis for the collection and classification of lotus germplasm resources.The results showed that nine SSR fluorescent labeling markers combined with capillary electrophoresis detection method were used to detect the polymorphism of 72 lotus samples.In total,51 polymorphic alleles were detected,with an average of 5.67 alleles per loci.The polymorphism information content PIC was ranged from 0.34 to 0.76,with an average of 0.56.72 lotus cultivars could be fully distinguished by using 6 pairs of SSR primers,including CL04,SSR449,SSR412,CL01,CL16 and CL10.SSR fingerprints of 72 lotus cultivars were established by combining all the amplified bands of each primer,the clustering dendrogram was further performed by using SSR fingerprint data,and the results showed that 72 samples could be grouped into three major groups at the similarity coefficient of0.535.This work will provide valuable information for the classification and molecular breeding of lotus cultivars in the future.
