不同运动方式对肥胖大鼠脂肪组织BMP-4和BMP-7及其相关产热调控蛋白表达的影响 被引量:5

Effects of Different Exercise Regimens on the Protein content of BMP-4/7 and Thermogenesis-related Proteins in Adipose Tissue of Obese Rats

摘要 目的:探讨不同运动方式对肥胖大鼠脂肪组织中BMP-4/BMP-7及其相关产热调节蛋白表达的影响,探寻促进肥胖大鼠脂肪产热代谢的有效运动类型及其调控机制。方法:从96只已经进行了8周高脂饲养的Sprague-Dawley大鼠中,筛选出体重最大的36只作为肥胖组大鼠,并将其随机平均分为肥胖安静组(n=10)、肥胖有氧运动组(n=10)和肥胖高强度间歇运动(HIIT)组(n=10),其余6只处死检测血脂水平。同时,从16只已经进行了8周标准饲养的同龄大鼠中随机筛选出10只作为标准安静对照组,其余6只处死检测血脂水平。肥胖有氧运动组以60%~70%VO2max强度在跑台上持续运动,肥胖HIIT组以70%—90%—50%—70%VO2max强度的跑速在跑台上做交替运动,每天50 min,5 d/周,共8周。第8周末,最后1次运动后48 h,取大鼠腹主动脉血、棕色脂肪、皮下和睾周白色脂肪。采用Western blot检测脂肪中BMP-4、BMP-7、TBX1、PRDM16和UCP-1的蛋白表达量。结果:1)肥胖有氧运动和HIIT组血清TC、TG和LDL-C水平低于肥胖安静组(P<0.05);2)与肥胖安静组相比,肥胖有氧运动组大鼠皮下脂肪组织BMP-4、BMP-7、TBX1、PRDM16和UCP-1的蛋白含量显著增加(P<0.01);棕色脂肪组织中BMP-4蛋白含量显著增加(P<0.01),BMP-7、UCP-1的蛋白含量显著降低(P<0.01)。3)与肥胖安静组相比,肥胖HIIT组大鼠睾周脂肪组织BMP-4、TBX1和UCP-1蛋白含量显著增加(P<0.01);棕色脂肪组织中BMP-7、UCP-1蛋白含量的显著降低(P<0.01)。结论:8周的有氧运动对肥胖大鼠皮下脂肪BMP-4和BMP-7蛋白水平而HIIT对睾周脂肪中BMP-4蛋白水平有更明显的影响,这2种运动对棕色脂肪的BMP-7蛋白表达有抑制作用。运动干预后,肥胖大鼠UCP-1的变化在皮下脂肪和棕色脂肪中与BMP-7的变化相关程度更高,而在睾周脂肪中与BMP-4变化的相关程度更高。 Objective:Elucidate the effects of a high-fat diet and different exercises on the expression of BMP-4/7 in adipose tissue.Method:Thirty obese male Sprague-Dawley rats(11 weeks of age),which were screened by weight from 96 rats fed with high-fat-diet for 8 weeks,were divided into three groups:control obese group(OB-C,n=10),aerobic-exercise obese group(OB-AE,n=10),and high-intensity-interval-training obese group(OB-HIIT,n=10).Ten same-week-old standard Sprague-Dawley rats,which were screened randomly from 16 rats fed a standard feed for 8 weeks served as control standard-diet group(SD-C).The aerobic exercise groups exercised on the treadmill at an intensity of 60~70%of VO2max,while the HFD-HIIT group alternated exercise on the treadmill at intensities of 70%,90%and 50%of VO2max;training intervention continued for 8 weeks,5 days a week,50 minutes a day.At the end of the 8th week of training,48 hours after the last exercise,abdominal aortic blood,brown fat,subcutaneous and peri epididymal white adipose tissue were taken from all rats.The protein expression levels of BMP-4,BMP-7,TBX1,PRDM16 and UCP-1 in adipose tissue were detected by Western blot.Result:(1)Serum TC,TG,and LDL-c levels were lower in excercise groups than in the control groups(P<0.05).(2)Compared with OB-C group,the levels of BMP-4,BMP-7,TBX1,PRDM16 and UCP-1 in groin-subcutaneous white adipose tissue of obese rats in OB-AE group increased significantly(P<0.01);the level of BMP-4 in brown adipose tissue increased significantly(P<0.01),while the level of BMP-7 and UCP-1 decreased significantly(P<0.01).(3)Compared with OB-C group,the BMP-4、TBX1 and UCP-1 protein levels of peridymal adipose tissue were significantly increased(P<0.01)in OB-HIIT group;while the protein levels of BMP-7 and UCP-1 decreased significantly(P<0.01).Conclusion:The effects of 8 weeks of aerobic exercise on subcutaneous fat BMP-4 and BMP-7 protein levels in obese rats and HIIT on BMP-4 protein levels in periphytic fat were more obvious,while the two types of exercise had an inhibitory effect on the BMP-7 of the brown fat.In adipose tissue of obese rats,the change of UCP-1 induced by exercise is more related to the change of BMP-7 in groin-subcutaneous white adipose tissue and BAT,and it is more closely related to the change of BMP-4 in peri-epididymal white adipose tissue.
