摘要
目的:研究苦豆子总碱(total alkaloid of sophora alopecuroides,TASA)对小鼠急性酒精性肝损伤的保护作用和作用机制。方法:实验小鼠按体重随机分为空白对照组、模型组、联苯双酯阳性对照组、TASA低、中、高剂量组,连续灌胃给药7 d,然后以50%乙醇制备小鼠急性肝损伤模型,检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平,计算小鼠肝脏指数,显微镜下观察肝脏病理切片。结果:与空白对照组相比,模型组小鼠ALT、AST和MDA含量升高(P<0.05),SOD活性下降(P<0.05),HE染色显示肝细胞索排列紊乱,肝细胞肿胀、炎症细胞浸润且呈现大小不一的空泡;给药组与模型组相比,TASA组小鼠血清中ALT、AST、MDA含量降低(P<0.05),SOD活性升高(P<0.05);HE染色显示TASA组肝组织病理学变化减轻。结论:TASA对酒精所致的小鼠肝脏损伤具有一定的保护作用,这可能与TASA能减轻氧化应激作用有关。
Objective:To study the protective effect and mechanism of total alkaloids of Sophora alopecuroides(TASA)on acute alcoholic liver injury in mice.Methods:experimental mice were randomly divided into blank control group,model group,positive control group,TASA low dose group,TASA middle dose group and TASA high dose group,and intragastric administration for 7 days,and then a mouse acute liver injury model was prepared by giving 50%alcohol.The main indicators measured were mouse liver index,serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),malondialdehyde(MDA),superoxide dismutase(SOD)levels,and liver pathological sections.Results:Compared with the blank control group,the ALT,AST and MDA contents in the model group were significantly increased,and the SOD activity was significantly reduced,HE staining showed disordered arrangement of liver cells,swelling of hepatocytes,infiltration of inflammatory cells,and vacuoles of different sizes;compared with the model group,serum ALT,AST,and MDA levels in the TASA groups were significantly reduced(p<0.05,p<0.01),SOD activity was significantly increased(p<0.01),the pathological changes of liver tissue were significantly improved.Conclusion:It is concluded that TASA has protective effect on liver injury induced by alcohol in mice,which may be related to TASA’s ability to reduce oxidative stress.
