烟草赤星病拮抗菌的筛选、鉴定及其培养基优化 被引量:6

Screening and identification of antagonistic bacteria against tobacco brown spot and media optimization

摘要 为获得能有效抑制烟草赤星病病菌的生防拮抗菌,采用平板对峙法进行菌株筛选,再用菌丝生长速率法和离体叶片试验法对拮抗菌的拮抗效果进行测试,然后通过形态、生理生化特征以及16S rDNA、gyrA序列分析等鉴定拮抗菌种类,并对其生长条件进行了优化。结果筛选出一株有一定抑菌效果的拮抗菌T11,其平板抑菌率达到45.9%,离体烟叶试验法平均防效为64.6%,无菌滤液对菌丝生长的抑制率达到60.6%;经鉴定T11菌株为枯草芽孢杆菌(Bacillus subtilis),最适生长的营养条件组合为牛肉膏5 g/L、蔗糖15 g/L、MgSO4·7H_(2)O15 g/L,该菌株及相关营养条件可为烟草赤星病生物防治提供参考。 In order to obtain antagonistic bacteria which can effectively inhibit Alternaria alternata.Plate confrontation method was used for screening,and mycelial rate method and in vitro leaf test were used to test the antagonistic effect of isolated strains.The antagonistic bacteria species were identified by morphological,physiological and biochemical characteristics,16 S rDNA and gyrA sequence analysis.Then,their growth media were optimized.An antagonistic strain T11 with certain inhibition to the mycelial growth was screened out.The plate inhibition rate was 45.9%,the average control effect of tobacco leaf in vitro test was 64.6%,and the inhibition rate on mycelial growth was 60.6%.The strain T11 was identified as B.subtilis.The optimized medium for the strain T11 was beef extract 5 g/L,sucrose 15 g/L,MgSO_(4)·7H_(2)O 15 g/L.It provides a new reference for biological control of tobacco brown spot.
