基于一体化进样系统的人体尿液元素的电感耦合等离子体质谱快速检测方法

摘要 目的建立人体尿液中18种元素的电感耦合等离子体质谱(ICP-MS)快速检测方法。方法取400μl尿液,加入2%硝酸的100μg/L Au稀释液4 ml,使用一体化进样系统(ISIS),样品在八级杆氦气碰撞反应池,通过动能歧视消除干扰,采用ICP-MS法达到快速检测尿液中18种元素的能力。结果尿液中18种元素的线性范围为0.1~1000.0μg/L,所得回归方程的线性关系良好,r≥0.99。该方法的检出限为0.003~0.96μg/L,定量下限为0.01~3.22μg/L,回收率范围为82.1%~110.4%,精密度范围为0.7%~30.5%。此外,检测效能由每小时25个样本提高到每小时52个样本。结论该方法前处理操作简单,进样速度快,灵敏度、准确度、重复性满足尿液元素检测的需求,尤其适用于筛查、队列研究等大批量尿液检测。 Objective To establish a rapid method for the determination of 18 elements in urine by inductively coupled plasma mass spectrometry(ICP-MS).Methods The urine samples(400μl)were added with 4 ml 2%nitric acid+100μg/L Au solution.After pre-treatment,the samples were injected into the integrated injection system(ISIS).As the samples entered octopole reaction system,the interference was eliminated by kinetic energy discrimination(KED)using helium as it's collision gas.The contents of 18 elements in urine were determined by ICP-MS.Results This method was linear for all elements(correlation coefficient≥0.99)in the range of 0.1-1000.0μg/L.All 18 elements in urine were detected with the limit of detection(LOD)ranging from 0.003μg/L to 0.96μg/L,limit of quantification(LOQ)ranging from 0.01μg/L to 3.22μg/L.The recoveries of 18 elements in urine ranged from 82.1%to 110.4%,and the RSDs ranged from 0.7%to 30.5%.In addition,the efficiency of testing was increased from 25 samples per hour to 52 samples per hour.Conclusion This method has the advantages of simple pre-treatment and rapid sample injection,and the sensitivity,precision,and repeatability meet the requirements of urine elements detection.Overall,the method is particularly useful for screening,cohort studies and other urine tests of large sample.
