氧化苦参碱对糖尿病肺损伤的保护作用 被引量:1

Protective effect of oxymatrine on lung injury in diabetic rats

摘要 目的:氧化苦参碱已被用于糖尿病并发症的治疗,而氧化苦参碱是苦参的主要活性成分,用于研究其对糖尿病大鼠肺组织损伤的影响。方法:本文采用MTT和流式细胞仪检测不同浓度氧化苦参碱处理的BEAS-2B细胞的增殖和凋亡情况。酶联免疫吸附试验,定量实时聚合酶链反应和HE染色、免疫组织化学法评价肺组织的炎症反应和病理变化。采用Western blotting检测肺组织PI3K和MAPK蛋白表达。结果:结果表明,氧化苦参碱对BEAS-2B细胞增殖和凋亡无影响。氧化苦参碱可抑制炎症反应,促进PI3K和MAPK信号通路的激活,防止高血糖诱导的肺组织损伤。结论:氧化苦参碱的保护作用表明氧化苦参碱是治疗糖尿病的潜在药物。 Objective Oxymatrine(OMT)has been used to treat diabetes,while oxymatrine is the main active ingredient component of Sophora flavescens Ait.This study was aimed to evaluate the effects of OMT on lung injury in diabetic rats.Methods In this study,MMT assay and flow cytometry were applied to detect the proliferation and apoptosis of BEAS-2B cells treated with different concentrations of oxymatrine.The inflammatory response and pathological change in the lung tissue of diabetic rats treated with oxymatrine were evaluated by ELISA assay,QRT-PCR,and HE staining.Meanwhile,PI3K and MAPK signaling pathway proteins in lung tissue were determined by Western blot analysis.Results The results showed that OMT had no significant influence on the proliferation and apoptosis of BEAS-2B cells.OMT can inhibit inflammatory response and promote the activation of PI3K and MAPK signaling pathways to prevent damages of lung tissues induced by hyperglycemia.Conclusion The protective effect provided by OMT indicates that oxymatrine is a potential drug for the treatment of diabetes.
