盐反应性肠道共调节Th17细胞在实验性自身免疫性葡萄膜炎发病中的作用 被引量:1

Role of salt reactive intestinal coregulatory Th17 cells in the pathogenesis of experimental autoimmune uveitis

摘要 目的探究高盐饮食(high salt diet,HSD)对肠道微生物-免疫系统的影响,以及其在实验性自身免疫性葡萄膜炎(experimental autoimmune uveitis,EAU)中的作用。方法将鼠龄6周的130只C57BL/6小鼠随机分为EAU组(67只)和对照组(63只),对照组小鼠正常盐喂养,EAU组只进行高盐喂养,建立EAU模型并进行EAU评分。在EAU小鼠造模后第21天高峰期,取小鼠视网膜,进行RNA提取并进行RT-qPCR检测及分析白细胞介素-17(interleukin-17,IL-17)、白细胞介素-23(interleukin-23,IL-23)及维甲酸相关孤儿核受体(retinoic acidrelated orphan receptor gamma t,RORγt)mRNA的相对表达情况。取小鼠引流淋巴结及脾脏组织,提取单细胞悬液并进行流式染色,流式细胞仪检测并分析EAU小鼠淋巴结及脾脏Th17细胞比例。结果与对照组比较,EAU组小鼠炎性症状加重(P<0.05)。高盐饮食促进了EAU小鼠视网膜中炎症相关因子IL-17(P<0.001)、IL-23(P<0.005)及RORγt(P<0.005)mRNA表达,并促进EAU小鼠外周免疫器官(淋巴结及脾脏)Th17细胞增加(P<0.001)。结论HSD可促进致病性Th17细胞的生成且Th17细胞在EAU中起致病性作用。 Objective To investigate the effect of high salt diet(HSD)on intestinal microbial immune system and its role in experimental autoimmune uveitis(EAU).Methods A total of 130 six weeks old C57BL/6 mice were randomly divided into EAU group(n=67)and control group(n=63).The control group mice were fed with normal salt,and the EAU group was only fed with high salt,then the EAU model was established and EAU score was performed.At the peak stage of the 21st day after the establishment of the EAU mouse model,the mouse retina was taken for RNA extraction,RT-qPCR detection and analysis of interleukin-17(IL-17),interleukin-23(IL-23)and RORγt relative expression of mRNA.The draining lymph nodes and spleen tissues of mice were taken,the single cell suspension was extracted and flow cytometry was used to detect and analyze proportion of Th17 cells in lymph nodes and spleen of EAU mice.Results The results showed that the inflammatory symptoms of EAU group mice were worse than those of the control group(P<0.05).High salt diet can promote inflammation related factors IL-17(P<0.001),IL-23(P<0.005)and RORγt(P<0.005)in retina of EAU mice.The expression of mRNA can promote the increase of Th17 cells in peripheral immune organs(lymph nodes and spleen)of EAU mice(P<0.001).Conclusion HSD can promote the production of pathogenic Th17 cells and Th17 cells play a harmful role in EAU.
