TRIP13通过Hippo信号通路促进非小细胞肺癌的增殖、迁移和侵袭 被引量:1

TRIP13 promotes the proliferation and migration of non-small cell lung cancer via Hippo signaling pathway

摘要 目的探讨TRIP13在肺癌中的作用和信号传导机制。方法通过基因转染和siRNA干扰调控肺癌细胞中TRIP13的表达量;采用克隆形成实验、划痕实验以及基质胶侵袭实验观察TRIP13对肺癌细胞增殖、迁移和侵袭能力的影响;采用Western blot检测TRIP13的表达量对Hippo信号通路蛋白的影响。结果检索GEPIA数据库发现,TRIP13在肺癌组织中的表达水平均明显高于癌旁正常肺组织(P<0.01);TRIP13的高表达与肺癌患者的低生存率正相关(P=0.0011)。TRIP13的过表达可显著促进肺癌细胞的增殖、迁移和侵袭能力,并显著上调YAP、RhoA、CyclinB1和MMP9的表达,抑制LATS、MST和p-YAP的表达(P<0.05);而下调TRIP13后,则逆转上述结果。结论TRIP13调控Hippo信号通路促进非小细胞肺癌的增殖、迁移和侵袭。 Objective To investigate the role and molecular mechanism of TRIP13 in lung cancer cells.Methods The expression of TRIP13 in lung cancer cells was regulated by gene transfection or siRNA interference.The effects of TRIP13 on the proliferation,migration and invasion of lung cancer cells were examined using clone formation assay,scratch assay,matrigel invasion assay.The levels of proteins in Hippo signaling pathway were examined by Western blot.Results The expression of TRIP13 was significantly higher in lung cancer tissues than in normal lung tissues based on the GEPIA database(P<0.01),and the high expression of TRIP13 was correlated with poor prognosis(P=0.0011).Overexpression of TRIP13 significantly promoted the proliferation and migration and invasion of lung cancer cells.TRIP13 overexpression significantly upregulated the expression of YAP and RhoA and Cyclin B1 as well as MMP9,but inhibited the expression of LATS,MST and p-YAP(P<0.05).After decreasing the expression of TRIP13,the above results were reversed.Conclusion TRIP13 promotes the proliferation,migration and invasion of NSCLCs via Hippo signaling pathway.
