苓桂术甘汤通过NLRP3/ASC/caspase-1途径改善过敏性哮喘小鼠气道炎症反应 被引量:2

Lingguizhugan Decoction ameliorates airway inflammation in allergic asthmatic mice through NLRP3/ASC/caspase-1 pathway

摘要 目的探讨苓桂术甘汤对过敏性哮喘小鼠炎症反应的影响及可能机制。方法40只SPF级雄性BALB/c小鼠随机分为空白对照组、模型组、苓桂术甘汤低、中及高组,每组8只。采用卵白蛋白(OVA)致敏联合雾化激发的方法建立过敏性哮喘小鼠模型。检测各组小鼠增强呼吸间歇(Penh)值并评估小鼠气道反应性;HE染色分析肺组织病理损伤;对支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中炎症细胞进行分类并计数;酶联免疫吸附法(ELISA)检测小鼠BALF中炎性因子IL-4、IL-5、IL-13、IFN-γ、IL-1β及IL-18表达;Western blot检测NLRP3、ASC、pro-caspase-1及cleaved-caspase-1蛋白表达。结果与模型组相比,苓桂术甘汤能降低过敏性小鼠Penh值;改善小鼠肺组织病理损伤;减少小鼠BALF中中性粒细胞、嗜酸性粒细胞及淋巴细胞数;降低BALF中炎性因子IL-4、IL-5、IL-13、IL-1β及IL-18的表达,增加IFN-γ的表达;抑制NLRP3、ASC、pro-caspase-1及cleaved-caspase-1蛋白表达。结论苓桂术甘汤能够抑制过敏性哮喘小鼠炎症反应,其作用机制可能与抑制NLRP3炎症小体通路之间存在一定的关联。 Objective To investigate the effect of Lingguizhugan Decoction on inflammatory response in mice with allergic asthma and its possible mechanism.Methods 40 SPF male BALB/c mice were randomly divided into blank control group,model group,and low-,medium-and high-dose group of Lingguizhugan Decoction,with 8 mice in each group.A mouse model of allergic asthma was established by using ovalbumin sensitization combined with nebulization.The enhanced respiratory interval(Penh)value of mice was detected and the airway reactivity of mice was evaluated.Lung tissue pathological damage was analyzed by HE staining.The inflammatory cells in bronchoalveolar lavage fluid(BALF)was classified and counted.The levels of IL-4,IL-5,IL-13,IFN-γ,IL-1βand IL-18 in BALF were analyzed by ELISA.The expressions of NLRP3,ASC,pro-caspase-1 and cleaved-caspase-1 were detected by Western blot.Results Compared with the model group,Lingguizhugan Decoction can reduce the Penh value of mice with allergic asthma,improve the pathological damage of lung tissue in mice,reduce neutrophils,eosinophils and lymphocytes in BALF of mice,reduce the expression of inflammatory factors IL-4,IL-5,IL-13,IL-1βand IL-18 in BALF,and increases the expression of IFN-γ,inhibit the expressions of NLRP3,ASC,pro-Caspase-1 and cleaved-caspase-1.Conclusion Lingguizhugan Decoction can inhibit the inflammatory response in mice with allergic asthma,and its mechanism may be related to the inhibition of the NLRP3 inflammasome pathway.
