摘要
HRP轴突逆行传送的方法用于周围神经研究的历史还不到10年。1971~1973年Kristen-sson和Olsson^([1,2])等将之用于舌下神经的研究;1975年Ellison和Clark^([3])着眼于自主神经的周围部分对HRP摄取状态的研究,将1%的HRP溶液分别注入于海猽的前眼房、肾上腺髓质、胃和回肠壁、心壁及颈上节等处,追踪节前神经元的所在,并在迷走神经结状节和胸上段的脊神经节内发现了标记细胞,为用低浓度HRP溶液追踪内脏传入途径开创了先例。后来,用HRP法进行周围神经的研究逐渐增多,但总的看来,对传出神经研究的较多,而对传入神经的研究较少。内脏的传入冲动随自主神经系向中枢传送的现象,早在Langley时代即已被人们所认识。
Thirteen young rabbits of about 0.5 kg body weight were used.To each animal 200μl of 1.0%horseradish peroxidase(Sigma TypeⅥ)solution was injected into the upper,or the middle,or the lower region of the bladder wall unilaterally.After a survival time of 2 days,the animal was sacrificed,and the spinal ganglia on the injection side were treated according to Mesulam’s method.The distribution of the HRP labeled cells were examined under microscope,and the results were as follows:1.Using HRP solution with a concentration as low as 1.0%,we could find clearly labeled cells appearing in the spinal ganglia on the injection side from Th-9 to Co-1,of which ganglia S-3,4,5 were the segments where the labeled cells were most numerous,and ganglia L-3,4,5 next.This fact shows that the afferent fibers of the bladder enter the spinal cord by two routes,i.e.,parasympathetic and sympathetic,and that the cells of the afferent fibers are diffusely distributed,while at the same time concentrated in certain segments.2.In ganglia S-3,4,5 and L-3,4,5 where the labeled cells were most and next most numerous,80%of the labeled cells were in the former,whereas 20%in the latter,indicating that the afferent route via the pelvic nerve is predominant over the sympa-thetic.In this respect the results of the 3 different injection regions were essentially the same.3.By counting the labeled cells in ganglia S-3,4,5 Separately we could find out that each ganglion was connected with all the 3 regions of the bladder,yet there was still more or less apparent segmental characteristics in these connections.Thus,in animals injected in the upper region the percentages of the labeled cells in ganglia S-3,4,5 were 60.39%,32.87%,and 6.74%respectively;but in those injected in the lower region it was reduced to 34.36%in ganglion S-3 but increased in ganglion S-5 to 15.30%.The result of animals injected in the middle region lies between the former two.4.The afferent fibers accompanying sympathetic nerve,however,entered the thora-cic and lumbar ganglia with the highest percentage(40~48%)in ganglion L-4.5.Among the sacral ganglia,it was found that the labeled cells of different sizes were so distributed as to show that different types of labeled cells were connected with certain respective regions of the bladder.The percentage of the larger cells,in cases inject-ed in the upper region was 23.5%while in those injected in the lower region it was only 2.14%.And the percentage of the smaller cells was 34.4%in animals injected in the upper region but in those injected in the lower region it was sharply increased to 82%.The labeled cells in lumbar ganglia were mainly small ones(about 80%),the medium-sized cells were about 20%,and the large cells were only scanty.
