摘要
本文报道人体子宫颈癌HeLa细胞系细胞的超微结构,及其在培养液中加入10,20μg/ml醋酸棉酚,作用24,48h后的变化状态。HeLa细胞在常态生长情况下,具有一般癌细胞的超微结构特点。在醋酸棉酚的作用下,这些细胞间的紧密连接未见发生改变。而在细胞内,线粒体首先发生变化,由肿胀、嵴断裂、空泡化,直至溶解成颗粒状物。棉酚对HeLa细胞的损害作用,具有剂量相关性和作用时间相关性。在棉酚浓度加大和作用时间增长时,可见除线粒体以外的其他细胞器的改变,包括溶酶体大量增多、核膜受损及核内异染色质改变等。扫描电镜下,可见微绒毛断落,细胞表面出现极不正常的泡状突起。本文还对棉酚损伤HeLa细胞的性质及其意义进行了讨论。
The present paper described the ultrastructures of HeLa cells and the changes:after treated with gossypol acetic acid.Our observations showed that HeLa cells possessed all malignant morphological characteristics.Besides,we have found three types of tight junctions between HeLa cells:first,the junction consists of microvilli or pseudopadia between cells.Second,the microvilli projects from one cell and contact with the surface of another cell,and the third,cells directly contact each other between cell membrane surfaces.Under the concentration of 20μg/ml gossypol acetic acid,the first event observable was the swelling and vacuolation of mitochondria,the breakage of it’s crest,and the disintegration of the whole mitochondria.As mitochondria is an important organelle with respect to the cell respiration and energy metabolism,so the damage of mitochondria might play a role to the inhibition of cell growth and lead to the death of the cell.Our results also showed that the action of gossypol was dose-dependent and time-dependent.As the duration of gossypol treatment prolonged,the damage of mitochondria and other organelles became more evident including the damage of nuclear membrane,and at the same time usually accompanying an increase in amount of lysosomes.Scanning electron microscope observation demonstrated that following gossypol treatment microvilli of the HeLa cells were broken and some irregular vesicles appeared on the cell surface.No ultrastructural damage in the Golgi apparatus and the tight junction between cells could be seen even under higher concentration(20μg/ml)of gossypol treatment for 48 hrs.The action and nature of gossypol damaged HeLa cells and its significance were discussed.
