3-氯丙二醇和2,3-氧丙醇小剂量合并长期间歇用药后某些器官的组织学检查

摘要 本实验每日以5mg/kg体重3-氯丙二醇和75mg/kg体重2,3-氧丙醇合并用药给雄大鼠灌胃,连续2天,停药8天,以10天为一疗程,共给药9疗程。于给药3、6、9疗程及3、6疗程停药后90天,9疗程停药后120天,分别对大鼠睾丸、附睾的组织学改变以及内脏器官组织学变化、末梢血液细胞计数及分类、骨髓细胞增生度及骨髓细胞微核率进行检查。研究结果表明,合并用药引起睾丸生精上皮功能抑制和附睾内精子退变所产生的抗生育作用是可逆的;对造血功能以及内脏的组织学结构、骨髓细胞微核率的影响不明显,或无统计学意义。因此,以小剂量3-氯丙二醇和2,3-氧丙醇间歇(疗程)长期给药,无明显毒性和副作用。 The complete antifertility action of alpha-ehlorohydrin in combination with glycidol with a daily oral dose of 5mg/kg and 75mg/kg for two days has been demonstrated by our previous studies.In this experiment the rats received 9 course treatments,one of which included two-day-administration with a daily dose mentioned above and eight-day-cessation of medication.The specimens of some inner organs(heart,liver,kidney,lung and spleen),blood and bone marrow were taken at the end of 3rd,6th,and 9th course treatment,and on the 90th day after cessation of medication of 3rd and 6th course treatment and on the 120th day after that of 9th course treatment.The mating test of treated male rats with untreated female started on the 90th day after cessation of medication of 9th course treatment.The complete inhibition of fertility was found at the end of each course treatment.But its recovery occurred in 70%of treated animals on the sampling day of cessation of medication.There was no significant changes of histo-pathology in the inner organs.The blood cell count and differential count,and the proliferation and the rate of micronucleus of bone marrow cells,were not significantly affected.The results suggest that a low combined dose of alpha-chlorohydrin and glycidol could be used twice every 10 days for a long-term without obvious side effects.
