人参皂苷对肺癌细胞增殖、凋亡影响及作用机制 被引量:6

Effect and mechanism of ginsenosides on proliferation and apoptosis of lung cancer cells

摘要 目的探讨人参皂苷Rg3对非小细胞肺癌细胞系NCI-H292细胞增殖、凋亡的作用及其与相关信号通路的关系。方法用不同浓度(0、20、40、60μmol/L)人参皂苷Rg3处理NCI-H292细胞。CCK8法检测人参皂苷Rg3对NCI-H292细胞增殖的影响;流式细胞术检测人参皂苷Rg3对NCI-H292细胞凋亡的影响;Western blot检测caspase-3和Bcl-2蛋白表达;反转录聚合酶链反应检测Keap1 mRNA和Nrf2 mRNA表达,观察人参皂苷Rg3对Keap1-Nrf2/ARE信号通路的调节作用;通过检测活性氧水平评价细胞内氧化应激水平。结果与对照组(人参皂苷Rg3为0μmol/L)比较,随着人参皂苷Rg3浓度增加,细胞的增殖能力和Nrf2 mRNA表达下降,细胞的凋亡能力、caspase-3和Bcl-2蛋白表达量、Keap1 mRNA表达、活性氧水平上升,且均呈浓度依赖性(P<0.05)。结论人参皂苷Rg3可能通过抑制Keap1-Nrf2/ARE信号通路而抑制非小细胞肺癌细胞增殖,并促进其凋亡。 Objective To investigate the effects of ginsenoside Rg3 on proliferation and apoptosis of NCI-H292 cell line of non-small cell lung cancer(NSCLC)and its relationship with related signaling pathways.Methods NCI-H292 cells were treated with ginsenoside Rg3 at different concentrations(0,20,40,60μmol/L).CCK8 assay was used to detect the effect of ginsenoside Rg3 on NCI-H292 cell proliferation.The effect of ginsenoside Rg3 on apoptosis of NCI-H292 cells was detected by flow cytometry.Western blot was used to detect caspase-3 and Bcl-2 protein expression.The expressions of Keap1 mRNA and Nrf2 mRNA were detected by reverse transcription polymerase chain reaction,and the regulation of ginsenoside Rg3 on Keap1-Nrf2/ARE signaling pathway was observed.The level of intracellular oxidative stress was evaluated by reactive oxygen species.Results Compared with the control group(ginsenoside Rg30μmol/L),with the increase of ginsenoside Rg3 concentration,cell proliferation and Nrf2 mRNA expression decreased,apoptosis,caspase-3 and Bcl-2 protein expression,Keap1 mRNA expression and reactive oxygen species increased,and all of them were concentration-dependent(P<0.05).Conclusion Ginsenoside Rg3 may inhibit the proliferation of lung cancer NCI-H292 cells and promote their apoptosis by inhibiting the expression of Keap1-Nrf2/ARE signaling pathway.
