不同产地红景天药材HPLC指纹图谱分析 被引量:5

Analysis on HPLC Fingerprints of Hongjingtian (Rhodiolae Crenulatae Radix Et Rhizoma) from Different Regions

摘要 目的采用高效液相色谱(high performance liquid chromatography,HPLC)分析不同产地红景天中药材的色谱指纹图谱。方法建立红景天药材的HPLC检测方法,固定相为Agilent C_(18)色谱柱,流动相才采用甲醇-0.05%磷酸溶液梯度洗脱(初始比例为1∶9),检测波长275 nm,流速为0.8 mL/min。以红景天苷为标示峰,西藏大花红景天为校正峰,建立西藏、四川、云南、吉林、新疆等不同产地红景天药材的色谱指纹图谱。结果对9个不同产地和品种的红景天进行多点校后,共标定9个共有峰。不同产地和品种的红景天共有峰的相对保留时间无明显差异,相对峰面积差异较为明显,其中西藏大花红景天的红景天苷和1、2、3、6、7、8、9色谱峰的峰面积均高于其它产地红景天。结论藏区大花红景天具有丰富的红景天苷及其它的共有成分,临床可根据红景天的HPLC指纹图谱进行鉴别和质量评估。 Objective To analyze chromatographic fingerprints of Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)from different regions by high performance liquid chromatography(HPLC).Methods HPLC test method for Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)was established.The stationary phase was Agilent C_(18)chromatographic column.The mobile phase was eluted with the gradient of methanol-0.05%phosphoric acid solution(initial ratio=1∶9).The detection wavelength was 275 nm and the flow rate was 0.8 mL/min.Taking rhodioloside as labeled peak and Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)as corrected peak,chromatographic fingerprints of Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)from Tibet,Sichuan,Yunnan,Jilin and Xinjiang were established.Results After multi-point calibration for different varieties of Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)from 9 different regions,9 common peaks were calibrated.There was no significant difference in the relative retention time among the common peaks of different varieties of Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)from different regions,while the differences in relative peak area were significant.The rhodioloside and peak areas of 1,2,3,6,7,8 and 9 chromatographic peaks in Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)were higher than those in Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)from other regions.Conclusion Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma)contains rich rhodioloside and other common components.Clinical identification and quality evaluation can be based on HPLC fingerprints of Hongjingtian(Rhodiolae Crenulatae Radix Et Rhizoma).
