摘要
目前冠心病在全球范围内仍是一个庞大的群体,严重危害人类生命和健康。三七总皂苷(tPNS)作为临床的常用处方药广泛应用于冠心病、高脂血症等血管性疾病的治疗中。目的该文将通过生物信息学预测三七总皂苷所调控的miRNA的标靶基因,并分析其所参与的生命过程,以探讨三七总皂苷通过miRNA治疗冠心病作用机制。方法通过中国知网检索三七总皂苷调控的miRNA,并通过mirDIP数据库(http://ophid.utoronto.ca/mirDIP/index.jsp)得到miRNA对应标靶基因,并有3个以上数据库验证标靶关系。经CooLGeN数据库(http://ci.smu.edu.cn/CooLGeN/Home.php)获得冠心病靶点。再利用Venny 2.1软件(http://bioinfogp.cnb.csic.es/tools/venny/index.html),获得miRNA标靶基因与冠心病靶点的交集。再将得到共同靶点在STRING平台(https://string-db.org/)构建PPI(protein-protein interaction)网络模型,明确蛋白相互关系。使用生物学信息注释数据库DAVID(https://david.ncifcrf.gov/summary.jsp)进行生物功能和通路分析。最后,利用Cytoscape 3.2.1软件,构建主要药物成份-miRNA-靶点-信号通路网络图。结果通过中国知网检索,得到三七总皂苷调控的主要miRNA有miR-let-7b、miR-21、miR-155、miR-34a,经过mirDIP数据库得到miR-let-7b、miR-21、miR-155、miR-34a对应标靶基因,再利用Venny 2.1软件获得miR-let-7b、miR-21、miR-155、miR-34a标靶基因与冠心病靶点的交集有341个,其中包括CAMTA1,PDCD4,IL6R,THBS1等靶点。经STRING平台得到蛋白相互关系。经过DAVID数据库,显示核心靶点与细胞凋亡过程、基因表达、炎症反应的正向调节等生物过程密切相关。结论三七总皂苷通过调控miRNA网络,进而影响多种蛋白质表达,参与炎症、凋亡等生命过程,以治疗冠心病。该研究结果为揭示临床应用三七类中药治疗冠心病的机制具有一定参考价值。
Objective At present,coronary heart disease is still a large group on a global scale,and it is a cardiovascular disease that seriously endangers human life and health.Panax notoginseng saponins(tPNS)are widely used as clinical prescription drugs in the treatment of vascular diseases such as coronary heart disease and hyperlipidemia.In this paper,we predicted the target genes of miRNAs regulated by tPNS by bioinformatics,and analyzed the life processes involved in them to explore the mechanism of action of tPNS on GI treatment of coronary heart disease.Methods The miRNAs regulated by tPNS were searched by Chinese Knowledge Network,and miRNA corresponding target genes were obtained through mirDIP database(http://ophid.utoronto.ca/mirDIP/index.jsp),and more than three database verified the targets relationship.Coronary heart disease targets were obtained from the CooLGeN databas(http://ci.smu.edu.cn/CooLGeN/Home.php).The Venny 2.1 software(http://bioinfogp.cnb.csic.es/tools/venny/index.html)was used to obtain the intersection of the miRNA target gene and the coronary heart disease target.A common target would be used to construct a PPI(protein-protein interaction)network model on the STRING platform(https://string-db.org/)to clarify the relationship among proteins.Biological function and pathway analysis were performed on common targets by using the biological information annotation database DAVID(https://david.ncifcrf.gov/summary.jsp).Finally,the main drug component-miRNA-target-signal pathway network map was constructed by using Cytoscape 3.2.1 software.The main microRNAs regulated by tPNS were microlet-7 b,microbe-21,microbe-155 and microbe-34 a.The microlet-7 b,microbe-21,microbe-155 and microbe-34 a were obtained from mirDIP database.The corresponding target genes were obtained by Venny 2.1 software.The microlet-7 b,microbe-21,microbe-155 and microbe-34 a were obtained by Venny 2.1 software.The target genes were related to coronary heart disease.There were 341 intersection targets,including CAMTA1,PDCD4,IL6 R,THBS1 and so on.The PPI was obtained by STRING platform.The DAVID database showed that the core targets were closely related to biological processes such as apoptotic process,gene expression and positive regulation of inflammatory response.The tPNS affect the expressions of various proteins by regulating the miRNA network,and participate in the life processes such as inflammation and apoptosis to treat coronary heart disease.The results of this study have certain reference value for revealing the clinical application of Panax notoginseng Chinese medicine in the treatment of coronary heart disease.
